Objective:Aleppo Gall is the main active ingredients extracted from Turkish galls, the mechanism of Aleppo Gall effected on oral bacterial is not clear. The aim of this paper is to study the inhibiting effect of Aleppo Gall on Streptococcus mutans in the state of planktonic and biofilm during the process of growth, acid metabolism and glycose metabolism; the effect of Aleppo Gall on lactate dehydrogenase (LDH) which is the main virulence factor lead to caries; on the activity of glucosyltransferase(GTF); and on the gene expression of gtfB,gtfC,gtfD. Then the paper also discusses the defence mechanism of Aleppo Gall on caries, provides theoretical and experimental basis for developing natural anti-caries preparation of Uighur. Methods:1) Slip diffusion method was used to measure the inhibition zone of each concentration of Aleppo Gall; two-fold dilution was used to measure Minimal inhibitory concentration (MIC) and Minimal bactericidal concentration (MBC) of Aleppo Gall effecting on the growth of Strepto-coccus mutans; The visible light semiquantitative method has been used to measure biomass (OD570) values so that the effect of medicine on biomass of oral bacteria biofilm has been discovered; 2) Using pH meter measureApH and Anthrone measured the effect of Aleppo Gall on acid ogenicity of Streptococcus mutans in different state and on intracellular polysaccharide; 3) Bradford method was used to measure the content of total protein of GTF,and anthrone method was used to measure gluean Polysaccharide. Assayed the activity of lactate dehydrogenase(LDH) with the pyruvate-dependent oxidation of NADH-with; 4)The gtfs mRNA levels under different condition were determined by real-time quantitative PCR. Results:1) Gallic tannins significantly inhibited the growth of Streptococcus mutans, MIC of gallic tanninson Streptococcus measured by dilution experiments mounted to 4mg/ml, MBC was 16mg/ml; the MBEC value of gallic tannin on the growth of single strain biofilm of Streptococcus mutans is 8mg/ml, which was measured by semi-quantitative determination of OD values in the condition of visible light; 2) Gallic tannins inhibited acid production of Streptococcus mutans and the synthesis of intracellular polysaccharide (WIG); 3) Results The activity of GTF and LDH in the state of biofilm was higher than planktonic; Aleppo Gall, NaF and Aleppo Gall union NaF inhibited GTF and LDH both in the state of planktonic and biofilm; LDH in the state of planktonic are more sensitive with drug. The activity of LDH was statistically significant in different culture conditions; the inhibition rate of Aleppo Gall is the highest, the secend one is NaF and the last one is Aleppo Gall union NaF; 4) Gallic tannins influenced the expression of Streptococcus mutans gtfs gene by different degrees. Conclusions:The effect of Aleppo gall and active components of Aleppo gall on oral Streptococcus mutans not only inhibits bacteria growth, acid metabolism and glycose metabolism, but also adjusts the expression of Streptococcus mutans gtfs gene, effectively prevent the occurrence of dental caries.
