Abstract/Summary:
Objective:To investigate the distributional characteristics of Adenosine Triphosphate binding cassette transport protein A1 (ABCA1) gene R219K polymorphism and association with blood lipid level in Chinese Han and Uigur population of Xinjiang. And the aim is to investigate the association between coronary heart disease (CHD) and R219K polymorphism of ABCAl gene and the influence of the level of serum lipid in Chinese han and Uighur population of Xinjiang. Methods:A total of 649 han with CHD and 640 health han peole were examined ABCA1 gene R219K polymorphism with polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) technique. The allele frequencies and genotypes of CHD group were compared with control group. A total of 471 Han with CHD and 468 health Han peole were examined ABCA1 gene R219K polymorphism with (PCR-RFLP) technique.The allele frequencies and genotypes of CHD group were compared with control group.The correlation of ABCAl gene R219K polymorphism and lipidemia was investigated.ResuIt:(1)The allele frequencies of ABCA1 gene R219K Were 0.21 and 0.14 in KK,were 0.51 and 0.52 in RK,were 0.28 and 0.34 in RR,0.46 and 0.40 in K,and 0.54 and 0.60 in R in Han and Uygur population, which showed significant difference (P<0.05). (2)The serum high density lipoprotein cholesterol(HDL-C) concentrations were higher and triacylglycerol (TG) levels were lower in Uygur and Han population with KK genotype than in those with RR genotype,which showed a significant difference (P<0.05). (3) The allele frequencies of ABCAl gene R219K Were 0.17 and 0.21 in KK,were 0.48 and 0.51 in RK,were 0.35 and 0.28 in RR,0.41 and 0.46 in K,and 0.59 and 0.54 in R in CHD and control in Han population, which showed significant difference (P<0.05). (4) The serum HDL-C concentrations were higher in CHD in Han population with KK genotype than in those with RR genotype as well as the serum TG and total cholesterol(TC) concentrations were lower in CHD in han population with KK and RK genotype than in those with RR genotype, which showed a significant difference (P<0.05). (5) The allele frequencies of ABCA1 gene R219K Were 0.13 and 0.14 in KK,were 0.42 and 0.52 in RK,were 0.46 and 0.34 in RR,0.33 and 0.40 in K, and 0.67 and 0.60 in R in CHD and control in Han population, which showed significant difference (P<0.05). (6) The serum HDL-C concentrations were higher in CHD in Uygur and Han population with KK genotype than in those with RR genotype,which showed a significant difference (P<0.05).Conclusions: (1)The frequencies of ABCA1 gene R219K genotypes and alleles in Han population are different from Uigur from Uigur population in xingjiang, The polymorphism may be associated with level of serum HDL-C and TG in Uighur and Han ethnic of Xinjiang. (2) The ABCA1 polymorphism of R219K gene has influence on level of serum HDL-C in Han and Uighur population of Xinjiang, which is associated with development of CHD. (3) K allele might be a protective factor of CHD.
