OBJECTIVE:Observes the effects of different dosage propofol on serum NO and NOS expression in tissues by intermittent intraperitoneal injection in rats.METHODS:24 male health Sprague-Dawley rats weighting 180g-220g were randomly divided into four groups(n=6),which were normal control group(A),10mg/kg dosage propofol group(B),20 mg/kg dosage propofol group(C),and 40mg/kg dosage propofol group(D). Rats were received with the dose of propofol by intraperitoneal injection every day. Ten days after administration, the levels of NO in serum and the activities of NOS in tissues of Myocardial,Kidney and Liver were measured by spectrophotometry. RESULTS:Compared with the A,B and C group, the levels of NO in serum and the expression of NOS in tissues of myocardial and kidney in D group were increased significantly (P<0.05). The remaining three groups showed no significant differences. Compared with the A group,the expression of NOS in tissues of livery in B,C and D group were significantly increased (P<0.05), and the three groups showed no statistically.CONCLUSION:The effect of intermittent high-dose intraperitoneal injection of propofol were increased the activity of NOS in tissues of Myocardial and Kidney, and increased the production of NO.
