Objective:Dental implants as a way of prosthodontics have gain their popularity with the progress in both technology and economic level nowadays. Bone growth factors are proved to be effective factors by researches focuing on the improvement of osseointegration of the bone and implants. however the defect vectors including the sources, the costs of bone growth factors,the immune response and side effects obstruct the clinical application of the exogenous growth factors. So an effective, economical, and convenient method promoting the growth of new bone around the implants and its osseointegration with the implants to ensure a successful dental implant is what dentists need urgently.This experiment studies the repair of the bone around the implants and its integration with the implants of rabbits after Xin Shang Xu Duan Tang (XSXDT) treatment. This will help us to explore the role and contribution of the traditional Chinese medicine treatment to the modern medicine.Method:32 male rabbits weighted about 3Kgs are divided into two groups randomly:the experiment group and the controlled group. Each group contains 16 rabbits. Dental implants were implanted into both sides of the ilium of the 32 rabbits. Each side has 4 implants. After the surgery the rabbits in the experiment group were ingested with XSXDT 15ml, qd, for 2 weeks while others in the controlled group were ingested with 0.9% NaCl instead. The all rabbits are phlebotomized 0.5ml venous blood from ear vein before surgery and 2,4,8,10,12 weeks after surgery to assay serum alkaline phosphatase, blood calcium and blood phosphorus.The rabbits(except the rabbit sacrificed at 2 weeks)have a subcutaneous injection with tetracycline hydrochloride solution at 2 weeks and 1 week before sacrificed. At 2 weeks,4 weeks and 8 weeks after surgery two rabbits in the experiment group and two in the controlled group are sacrificed respectively. The rest rabbits are sacrificed at 12 weeks after surgery.256 bone blocks containing the implants were cut to observe the osseointegration of implants and bone surrounding by whole specimen, X ray, fluorescence and SEM.The results are analyzed by the software spss 16.0.Results:1. Serum alkaline phosphatase, blood calcium and blood phosphorus:the content of serum alkaline phosphatase of the experimental group reached an earlier and higher peak than the controlled group. At 10-week afteroperation, the ALP of the experimental group almost descented to a level equal to the preoperation level. While, the ALP of the control groups didn't until 12-weeks afteroperation. The blood calcium level both of the experimental group and the controlled group step-up at 2- week afteroperation. And the blood phosphorus level descended. After that, the blood calcium level of both groups descented.2. General observation:The surgical wound of all animals healed well. All the implants connected firmly with the bone surrounding. No loosening or dropping were observed. We observed only very lamellar fibrous tissue on the specimen of 2-week afteroperation. 3. X-ray observation:Density of the bone around the implants of the experimental group was higher than the control group at 4-week afteroperation. The implants connected firmly with the bone at the experimental group at 8-week afteroperation, meanwhile, in the control group small transmission area could be observed.4. Fluorescence observation:The bone trabeculaes in the experimental group were more stronger and close-up at 4-week afteroperation. At 8-week afteroperation we could find the calcification of the new continuous bone trabeculae. Which was more obvious in the experimental group than in the control group. At 12-week afteroperation there are two continuous circular band of light around the implants. And in the experimental group the distance between the bands of light was wider than in the control group.5. SEM observation:In 2-week,4-week,8-week specimens there are gaps between the bone and the implants both in the experimental group and in the control group. In 12-week specimen, the bone and the implant closely integrated in the experimental group. But in the control group small space which was apparently be observed at 2000 times magnification can still be found between the bone and the implant.Conculsion:Xin Shang Xu Duan Tang can promote the proliferation of osteoblasts, speed up the process of bone regeneration. Thus it can improve osseointegration around the implants.
