Objective:Establish the rabbit model of lumbar disc degeneration, to observe the effect of traditional Chinese medicine Yao Bi Shu on early treated degenerated lumbar intervertebral disc of rabbit by X-ray imaging,histology morphology result and the impact of lumbar intervertebral disc in the proinflammatory factor IL-1αcontent and the expression of Sox9 mRNA. Preliminary study the molecular mechanism of this traditional Chinese medicine on degenerative disc.Material and method:Checking 54 healthy rabbits with similar species, weight and age. Rabbits were randomly divided into 3 groups (A:Yao Bi-Shu group, B:control group, C:normal group). Group A,B make model of lumbar disc degeneration and give each group different treatment.2,4,8 weeks after the operation, each group randomly selected three to be filmed at lumbar with X-ray, then Killed and taken L4/5 disc, perform anatomical observation, paraffin section and HE staining. After model succeed in 8 weeks, we treated group A with Yao BiShu, the other two group normal feeded. In 2,4 weeks, randomly select 4 models from each group, remove degenerated lumbar intervertebral disc and separately test the content of proinflammatory factor IL-1αusing ELISA in each group disc and the expression of sox9 mRNA using quantitative real-time PCR. The results use SPSS 13.0 software package to analyze variance (F test), multiple comparison, and T-test, significant level of a=0.05. Result:1. Establishment of rabbit degenerative disc model:Compared with normal group, control group rabbit models' vertebral filmed with X-ray. Image of disc space, the edge of vertebral of control group rabbit model show highdensity video; We can see at the edge of vertebral a large number of bone superfluous form in anatomy; Disc tissue HE stained show that the structure of annulus fibrosus damaged, there are types of cartilage formation and ossification in nucleus pulposus.2 Control group IL-1αlevels is the highest in all groups. IL-1αin Yao Bi Shu Group disc tissue compare to normal control group, higher. Intergroup difference was significant (P<0.05),while compare to the control group were significantly different (P<0.05).3 The expression of sox9 mRNA in Yao Bi Shu group are more than that in control group, intergroup difference was significant (P<0.05).4 The result of correlation analysis in Yao Bi Shu Group between IL-1αcontent and the expression of Sox9 mRNA is negative.Conclusion:1. We succeed in establishing rabbit lumbar intervertebral disc degeneration model induced by Annulus fibrosus puncture and injection, and the change of imaging and organization structure similar to the human lumber disc degeneration.2. Inflammatory cytokines IL-1αin the degenerated rabbit lumbar intervertebral disc can be induced by Yao Bi Shu treatment.3. Yao Bi Shu can increase expression of Sox9 mRNA.4 Yao Bi Shu can drive the expression of Sox9 mRNA due to inhibit the activity of IL-1α.
