| Objective:To investigate the influence to the proliferation and its differentiation to tendon cells of ADSC when treated with tendon tissues extracts, then provide a scientific basis for tendon tissue engineering and its ultimate clinical application.Methods:3 weeks old SD rats, isolate adipose tissue from its inguinal fat pads, then isolate stem cells by Collagenaseâ… digestion, seeded the stem cells in DMEM medium which contains fetal bovine serum for separation and subculture. Observe the cell morphology and growth by inverted microscope daily, MTT method to draw ADSC growth curve. After identified, take the third generation of ADSC and divide it into the experimental group and the control group, the exprimental group is treated with tendon extracts and the control group with Hank's solution, continue to culture and observation. MTT assay to compare the proliferation activity, typeâ… collagen immunocytes staining to test the differentiation into tendon and PT-PCR to test the expression of related genes (Collagenâ… , Collagenâ…¢gene).Results:4-6 hours after seeded, attachment were visible, it blooms after 7-8 days, arranged in a certain direction, present a swirl or a bundle, cells grew rapidly and stably after passage. After induction into the tendon, typeâ… collagen immunocytes staining showed that:the experimental group was positive while the control group was negative. PT-PCR showed that the expression of tendon related genes increased obviously (p<0.01). MTT experiments suggested that after the intervention of tendon tissues extracts, the proliferation activity of ADSC did not change, there's no significant difference compared to the control group (p>0.05).Conclusions:1. Tendon tissues extracts can not change the proliferation activity of rat ADSC.2. Tendon tissues extracts can promote rat ADSC differentiate into the tendon cells.
