BackgroundAs an interdisciplinary technology,tissue engineering is a combination of engineering and life sciences.Its main function is to restore,maintain and improve organizational functions through the development of biological substitutes.Tissue engineering technology develops very rapidly and is involved in various medical disciplines.It is widely used in the field of life sciences.Currently,in the field of oral surgery,periodontics,dental pulpology,implants and so on,all rely on the research progress of tissue engineering.Periodontitis is a disease with a very high incidence,which has a great impact on oral tissue damage,even in serious cases.Tissue defect,and when the oral cavity is in the state of periodontitis for a long time,plaque formation and cytokines,as well as inflammatory factors blocked by inflammation stimulation,will flow through the whole body,causing diseases of various systems,such as respiratory diseases,digestive tract diseases,kidney diseases,type 2 diabetes mellitus,premature birth,low birth weight newborns,rheumatoid arthritis.Alzheimer's disease and so on,so the treatment of periodontitis is the most urgent.At present,there is an urgent need to develop new and effective methods to repair tissue defects and promote tissue regeneration in the treatment of periodontitis.Traditional treatment methods can not achieve tissue regeneration,so tissue engineering technology is needed to achieve tissue regeneration.Tissue regeneration requires the coordination of growth factors,proteases,progenitor cells and immune cells.Mesenchymal stem cells replace adult cells,secrete growth factors and cytokines,and attract progenitors involved in tissue repair.Cells play an irreplaceable key role in tissue regeneration.In the process of tissue repair,because inflammation has a great impact on the repair process,we speculate that inflammation may also affect the regeneration function of mesenchymal stem cells,including migration,proliferation and differentiation.Therefore,we studied the changes of adipose-derived mesenchymal stem cells(ADMSCs)proliferation and differentiation in inflammatory environment.Objective:By studying the effects of inflammation on the proliferation and differentiation of adipose-derived mesenchymal stem cells,we can provide more evidence for the clinical application of adipose-derived stem cells and provide theoretical basis for further study of adipose-derived mesenchymal stem cells.Method:1.The rat model of chronic periodontitis was established by tethering.After 4 weeks,the inguinal adipose tissue was taken and adipose stem cells were extracted.2.Flow cytometry was used to identify the molecular phenotype of adipose-derived stem cells.3.were used to detect the expression of adipose stem cells(ADSCs)in inflammatory microenvironment.4.CCK-8 was used to detect the effect of inflammation on the proliferation of adipose stem cells.5.The effect of inflammation on the migration of adipose stem cells was investigated by scratch healing experiment.6.To observe the changes of differentiation function of adipose-derived stem cells by adipogenic and osteogenic staining experiments.Result:1.The alveolar bone absorption in the tethered group was observed after 4 weeks compared with that in the Untied group.2.Adipose stem cells were detected by flow cytometry.3.The results of qPCR and Western blotting showed that the expression of osteogenic genes ALP and RUNX2 in adipose mesenchymal stem cells of inflammatory rats was lower than that of normal rats(P < 0.05)..4.CCK-8 test results showed that compared with normal adipose stem cells,the growth rate of adipose stem cells in inflammatory rats was significantly faster(P < 0.05).5.Scratch test showed that the migration rate of adipocytes in inflammatory rats was significantly faster than that in normal rats.6.Inducers induce adipose mesenchymal stem cells to differentiate into osteoblasts.Calcium salts deposit on the surface of osteoblasts to form calcium nodules.Alizarin red staining shows that there are more alizarin red calcified nodules in the staining area.Standard inducers induce lipid droplets in adipose mesenchymal stem cells to differentiate into small round or round-like adipose stem cells.Fat droplets fused into large fat droplets.After oil red O staining,more brown red staining was found in oil red O staining area.Conclusion: Compared with normal rat adipose mesenchymal stem cells,the proliferation and migration of adipose mesenchymal stem cells in inflammatory rats increased,but the osteogenic differentiation ability decreased.
