ObjectiveTo observe the feasibility of constructing the analogue composite skin with soft tissue expander in vivo .MethodsThe soft tissue expander with two injection pots was implanted subcutaneously in the back, on both sides symmetrically in each of 10 rabbits and enlarged to 10ml by the injection pot A;1w after the surgery, fibrous capsulae formed around the expander and cultured analogue isogeneic rabbit epithelial cell suspension was injected subcutaneously by the injection pot B into the room between fibrous capsulae and expander . A control group was set up and the same volume normal saline was injected to the expander. 2w after injected the epithelial cell, the expander was removed to see whether the epithelialization could be found on the surface of fibrous capsulae by macroscopic and microscopic observation. That is whether the analogue composite skin whose structure is similar to the skin can form by combining fibrous cyst and injected epithelial cell.ResultsThe whole fibrous capsulae can form in the treatment group and the control group, easy to harvest.By comparing, transparency of cyst in the treatment group was slightly lower but thicker than of the control group by macroscopic observation, and no other significant difference was found .There was no epithelialization on the surface of fibrous capsulae in the control group by microscopic observation whereas structure similar to the epithelializatified on the surface of fibrous capsulae in the treatment group could be observed and covered the whole fibrous cyst tightly.ConclusionThe analogue composite skin in vivo whose structure is similar to the skin can be constructed with the above, before the ideal composit skin comes out from tissue engineering, the analogue composite skin constructed in this method may become the wound repair material of a major area burn and wounds.
