Background and objectiveAlthough many methods such as surgery retardation, drug retardation, growth factor therapy and gene therapy have been used to increase the skin flap survival, Partial necrosis of the skin flap, an important problem in plastic surgery, still needs further study. VEGF, the powerful growth factor of the endothelial cell, is the most important inductive factor of the blood vessel growing. Many researches have been proved that exogenous VEGF can stimulate the angiogenesis and increase the survival of skin flap and myocutaneous flap, but its effect is transient. Gene therapy with viral vector as the medium provides high expression efficiency and produces more effective and persistent function. Based on the above, this research adopts Ad-VEGF to inject into the expanded skin flap in white SD rats, then observed the change of the skin flap survival and morphologic change of the tissue.Materials and methodsRegardless of male or female, 20 white SD rats (weight: 300-490g), had been separated into two groups randomly (with 10 in each).
