ObjectiveAccording to the experiment on the streptococcus pneumoniae-infected aged rats and observe the influence that the mixture has made on the level of TNF-a,IFN-γ.,IL-8 and other agents on those rats' lungs, to explore the functional mechanism of the Sha Bai Pneumonia Mixture in treating the chronic pneumonia on aged patients.Methods1,To select 20 young and 60 aged SD male rats respectively. According to their weight, divide them randomly into the young blank control group, young model group, aged blank control group, aged model group, high-dose group and low dose-group of the Sha Bai Pneumonia Mixture and the group of Roxithromycin.2,To make an tracheotomy experimental model, inject bacterial liquid directly into trachea by micro-syringe, which can let rats be infected by the streptococcus pneumoniae three times a month to prolong its course of pneumonia.3,During the experiment, record the variation of those rats'weight, activity degree, food intake and other general cases.6 days after those rats taking such mixture, kill them and take out the lung tissues. Then, to homogenate a part of the tissue on the one hand, take out the serum and use ELIAS to measure the level of inflammatory factors, such as TNF-α,IFN-γ. On the other hand, to stain the other part of the lung tissue by HE, make slices of the pathological lung tissue and identify the pathological injure.Results①Apply non-parameter test to compare the aged group with low dose Chinese medicine group, the aged group of western medicine, and the aged group with high dose of Chinese medicine, it has statistical significance (P<0.05)②The comparison of the IFN-y level, between the aged group of western medicine, the aged group with high dose of Chinese medicine and the group with the low dose of Chinese medicine group does not have any Statistical significance.③The lung tissues of the aged and young model groups are both injured and the former group is more serious. As the Chinese and West medicine can protect the tissue, the impairment of lung tissues in the groups with Chinese drugs and western medicine is mild. ④In the lung tissue of pneumoniae-infected rats, expression of IL-8 is stronger, and its level is proportional to the severity of pneumonia; the group of the Sha Bai Pneumonia Mixture is same to Roxithromycin group, both of them can regulate the IL-8 of lung.ConclusionsThe Sha Bai Pneumonia Mixture has an apparent function to regulate the inflammatory factor on lung tissue of those Streptococcus pneumoniae-infected aged rats, It is achieved by the following reasons. First, the mixture can lower down the level of inflammatory factors, such as TNF-α,IFN-γ. Second, it can protect the injured lung tissue. Third, it can improve the weight of those Streptococcus pneumoniae-infected rats and also promote the nutrition state.
