In our country, coronet from Cervus nippon Temminok used as medicine had thousand years, the proteins and polypeptides are abundant in coronet. In this research, we used hight-press extract protein from coronet, determine the optimum extraction and preparation the coronet polypeptide by enzymatic hydrolysis.At last, researched the antioxidant and the antibacterial activity of it.1,Used coronet as material extraction as index, determine the optimum extraction by single factor and orthogonal experiment,the optimum extraction was:press was 0.10MPa,time was 15min,solid-liquid ratio was 1:5, and the extraction was 92.03%.2,Choose the trypsin and alkali protease as the best enzyme from neutral protease, papain, trypsin, pepsin and alkali protease.3,Used hydrolysis degree and scavenging action to hydroxyl radical as index, determine the optimum enzymolysis by single factor and orthogonal experiment, the optimum enzymolysis of trypsin was:pH 8,5, time was 5h, solid-liquid ratio was 5%, temperature was 50℃; the optimum enzymolysis of alkali protease was:pH 10.0, time was 5h, solid-liquid ratio was 4%, temperature was 50℃.And the optimum enzymolysis of double enzyme composite was:trypsin and alkali protease successively hydrolysis, pH 9.0 and 10.0, time was 5h for each, solid-liquid ratio was 5% and 6%, temperature was 55℃4,Research the antioxidant activity of polypeptide from coronet,the result showed the polypeptide can scavenge free radical,and the result of polypeptide was better than unhydrolysis protein.The IC50 of polypeptide scavenged hydroxy radical was 0.35 mg/mL; it was not so strong to DPPH, the IC50 of polypeptide scavenged DPPH was 12.05mg/mL, it's still need more hydrolysis; the IC50 of polypeptide scavenged O2-·was 0.63mg/mL.The polypeptide had ability to reductive Fe3+, presenting concentration-response relation and better than unhydrolysis protein; ability of resist lipid peroxidation was better than unhydrolysis protein, the IC50 was 0.59mg/mL.5,The polypeptide from coronet had bacteriostasic activity,and it's better than unhydrolysis protein,the activity changed by concentration of polypeptide.Two samples' diameters of bacteriostatic circle were 9.70mm and 8.00mm, they had bacteriostasic activity, but not best.
