Objective:To investigate the Influence of fluid shear stress on BMP2 and Smad4 of BMP2 signaling pathway in MG63 cells.Methods:Through fluid shear stress on MG63 osteoblastic cells in vitro 12dyn/cm20,15,30,60 and 120 minutes respectively, We used double-label immunofluorescence method to detect the expression levels of BMP2 and Smad4 in MG63 cells under different times of FSS. Statistical analysiswas performed using SPSS13.0 software package for one-way ANOVA.Results:Under 12dyn/cm2 of FSS, the expressions of BMP2 and Smad4 were significantly higher than those at 0 minutes groups(F=13.532, P=0.000; F=9.058, P=0.001)and affect maximal at 60 minutes.There was significant difference in expression of BMP2 in all groups (P<0.05). There was significant difference in expression of Smad4 in the 0,15,30 and 120 minutes groups (P<0.05). But in the 60 minutes groups,there was no significant difference in expression of Smad4 (P>0.05). Conclusion:BMP2 signaling pathway is an important pathway to the FSS, the appropriate level of FSS induced the expressions of Smad4 accumulation in cytoplasm through the higher level of the expressions of BMP2, Smad4 with RSmads complex, travel to the nucleus and act as activators and repressors of transcription of target genes.
