Objective:To investigate the effects of DDP at different concentrations on the inhibition of human cervical cancer SiHa cell proliferation and the expression of TSLC1 gene.Methods:After treatment at different concentrations of DDP (0-25ug/ml) for 24h,48h,72h. using MTT to determine the effects on SiHa cell proliferation, then measured OD, and calculated the rate of inhibition. The changes of expression of TSLC1 mRNA were detected by RT-PCR.Results:1. There are expressions of TSLC1 gene in human cervical cancer SiHa cell;2. With time of treatment or dose of DDP increasing, the inhibitory effect of DDP on the extracorporeal proliferation of SiHa cell was more evident(P<0.01).3. Intervention in different concentrations of cisplatin in human cervical carcinoma cell line SiHa different time. TSLC1 mRNA significant expression of the Increased (P<0.05), and showed significant measurement-time-dependent increase.Conclusion:1. There are expressions of TSLC1 gene in human cervical cancer SiHa cell;2. DDP could obviously inhibit the extracorporeal proliferation of SiHa cells by up-regulation of TSLC1 mRNA expression.
