Background and ObjectiveLung cancer is a most harmful malignancy to human health and life around the world, and the morbidity and mortality rates of it increased steadily and become a trend in the worldwide. In 2000,10 million new cases of Global cancer occurred,6.2 million cases were death cases, new cases of lung cancer of which were 1.239 million cases, accounting for 12.39%,1.1 million death cases, accounting for 17.7%. According to the domestic survey, the incidence rate of lung cancer increased by 26.9% each year, the mortality rate of it accounts for 22.7% of the total of cancer mortality, which has become the leading cause of cancer death. Non-small cell lung cancer accounts for about 80% of all lung cancer, in-depth study of its occurrence and control to reduce the overall morbidity and mortality of lung cancer is of great significance. Lung cancer is a multi-step, multi-stage, the result of multiple gene interactions, involving the activation of multiple oncogenes and tumor suppressor gene inactivation, in which tumor suppressor gene inactivation in the research is receiving increasing attention in recent years. TSLC1 (tumor suppress or in lung cancer - 1) is identified as a tumor suppressor gene through the complementary method in lung adenocarcinoma cell line A549,belongs to Necls family.TSLC1 Positioning on the human chromosome 11q23.2, containing 10 exon, are members of the immunoglobulin superfamily cell adhesion molecule encoding cell adhesion molecule in IgCAM, the molecule is a cell membrane proteins involved in interactions between cells. Studies have shown that most normal tissues outside the human body except for peripheral blood lymphocytes express TSLC1 molecules, it is mediated through homogeneous or heterogeneous between cell adhesion and in suppressing the occurrence of cancer play an important role. The current study found that loss or reduction of TSLC1 expression in the development of many cancers, including breast cancer, esophageal cancer and cervical cancer.4.1B(differentially expressed in adenocarcinoma of the lung) is found by the U.S. scientists TranYK in 1999 by differential display PCR technique (DDPCR), he found that loss of expression in lung adenocarcinoma and normal lung tissue with the expression of a gene, it belongs to the superfamily of proteins, Locating on human chromosome 18P11.3 region,4.1 protein interacts with actin, spectrin protein family of proteins and the cytoplasmic membrane protein, to maintain normal cell morphology and physiological characteristics. Studies have shown that 4.1B in the digestive system not only can maintain its normal structure but also can maintain the normal role of cell proliferation and adhesion, which indicated that loss of expression of the gene may weaken the connections between cells which can cause easy shedding of the tumor cells, it may be an important molecular basis of tumor metastasis.In current research, both of the genes are more studied in protein levels, and the correlations of them in non-small cell lung cancer are reported less in China. In order to provide for the transfer of non-small cell lung cancer:a clinical diagnosis, prediction and molecular therapy provide a theoretical basis. In this study, TSLC1, 4.1B expression in non-small cell lung cancer are detected by applying RT-PCR method from the gene level and the correlation between them and their relationship of clinical and pathological features of patients are analyzed.Materials and method1. All specimens of the Experimental group are from the Second Affiliated Hospital of Zhengzhou University, Thoracic Surgery, December 2009-2010 in April during the surgery and pathologically confirmed non-small cell lung cancer 52 cases of fresh tumor tissues and corresponding adjacent normal tissue 52 cases. Of which 33 cases were male,19 females; aged 35-73 years, mean 55.12±8.61 years; histological type:29 cases of squamous cell carcinoma,23 cases adenocarcinoma; differentiation:well differentiated in 17 cases,23 cases of moderate differentiated, poorly differentiated 12 cases; TNM staging:Ⅰof 15 cases,Ⅱof 24 cases,Ⅲof 13 cases. All patients were not carried out radiotherapy, chemotherapy and other treatment for the tumor biology before the surgery.2. By RT-PCR method, the expression of two genes 4.1B and TSLC1 are detected in 52 cases of non-small cell lung cancer and 52 cases of cancer and the adjacent normal lung tissue. Combined with clinical cases of patients'characteristics, the expression of the genes 4.1B and TSLC1 of 52 patients of tumor tissue and the relationships between characteristics of clinical cases are analyzed.3. All data are processed by SPSS11.5 statistical software, the data are expressed as x±s, quantitative data of two groups was used to compare with the t test, t'test are adopted where t test does not meet the conditions. Quantitative data was used to compare multiple sets of single-factor analysis of variance (one-way ANOVA), comparison between groups by LSD-t test. Pearson correlation analysis are performed to solve the correlation between indicators. Significance level is P<0.05.Result1. The expression of TSLC1mRNA in Carcinoma tissue was significantly lower than in adjacent normal lung tissue (0.349±0.008 vs 0.555±0.010) (P<0.01)。2. TSLC1mRNA in high differentiation group was significantly higher than that in poorly differentiated group (0.378±0.048 vs 0.345±0.061 vs 0.317±0.032) (P<0.05).TSLC1mRNA expression is related to the TNM staging of lung cancer, the later stages the less of the expression, phaseⅠ>phaseⅡ>phaseⅢ(P<0.05) The expression of TSLC1mRNA has nothing to do with Gender, age, histological classification (P>0.05)3. The expression of 4.1BmRNA in Carcinoma tissue was significantly lower than in adjacent normal lung tissue (0.209±0.040 vs 0.721±0.071) (P<0.01) 4.4.1BmRNA in high differentiation group was significantly higher than that in poorly differentiated group (0.233±0.032 vs 0.210±0.038 vs0.173±0.029) (P<0.05).4.1BmRNA expression is related to the TNM staging of lung cancer, the later stages the less of the expression, phaseⅠ>phaseⅡ>phaseⅢ(P<0.05).The expression of 4.1BmRNA has nothing to do with Gender, age, histological classification (P>0.05)5. Pearson correlation analysis showed:The expressions between TSLC1mRNA and 4.1BmRNA in Carcinoma tissue are proportionate (r=0.471,p<0.001).Conclusion1. TSLC1,4.1B expression in cancer tissues was significantly lower than in adjacent normal lung tissue.2. In Non-small cell lung cancer, the expression of TSLC1,4.1B has nothing to do and patient gender, age, pathological type; but with the non-small cell lung cancer differentiation, TNM staging, non-small cell lung cancer may reflect the degree of malignancy.3. Non-small cell lung cancer TSLC1mRNA expression is positively correlated with the 4.1BmRNA.4. TSLC1,4.1B are expected to be the target genes for diagnosis and therapy of non-small cell lung cancer gene.
