Ginseng lives in the northeast mountain area with the weather cold and wet. In the long process of its life, ginseng was vulnerable to various kinds of diseases.The stem was the most important transfusion and supportive tissue of the ginseng, meanwhile, ginseng stem has the similar pharmacology utility as its main root. Researches show that ginseng stem has different pathogenesis regularity with the change of the season. Also, the content patterns of the pharmacology utility were closely linked with the change of the season. However, the internal mechanism of the pathopoiesis and the synthesis rule active constituent were not clear. Protein is the direct regulators of the life activities and law and the ultimate expression results of the total gene subjected to various comprehensive factors, so the proteomics can be used to study the life activities and law between ginseng stems with different seasons directly and effectively.Ginseng stem contains high levels of contaminants such as celluloses and pectin, which are known to contribute to lower protein concentration and content. In this study, high-quality 2-DE pattern was obtained by means of optimization of protein extracted buffer and the method of removing impurity. And then, the 2-DE patterns of spring and autumn were compared to fix the differences protein points which were identified through MALDI-TOF-TOF. The mass spectrometric datas were retrieved by NCBInr database to fix the function of the protein points. In the end, we found that the protein with the energy metabolism and synthesis of growth substance contain large proportion among the protein that increased significantly in quantity in the stem of the spring season, and in the stem of antumn season, proteins resistance to adverse situation contain high levels among among the protein that increased significantly in quantity. The resuilts show that the stem of spring ginseng runs rapidly in cell division and growth and the stem of antumn grows slowly. However, the proteins against adversity were less than the antumn stem, which may be related to the pathogenic rule of spring and antumn stem of ginseng. Meanwhile, two proteins that were related to isoflavone and phenyl propane-liked compounds increased significantly in quantity, which may prompt spring is the best season to utilize these two substances.To sum up, this paper has studied the regular changes of ginseng life through the method of proteomics, which may lay the foundation for the study of ginseng physiological and pathological mechanism and promote the development of ginseng planting.
