PurposeThe human gene cysteine-rich 61 (Cyr61,CCN1), first cloned member of the CCN family, located on chromosome Ip22-p31. Purified Cyr61, which could support cell adhesion, promote cell migration, enhance growth factor-stimulated mitogenesis and play a role as a ligand of integrin receptor, was relevant to the genesis and metastasis of diverse carcinoma. But the level it expressed and the role it played in nasopharyngeal carcinoma remained unclear. This study detected Cyr61 expressed in the tissues of nasopharyngeal carcinoma and nasopharyngeal chronic inflammation patients, and aimed at figuring out the association between Cyr61 and nasopharyngeal carcinoma. Meanwhile, this study compared the expression level of Cyr61 among different types of syndrome in traditional Chinese medicine(TCM), as to explore the relevance between Cyr61 expression level and different types of syndrome in TCM.MaterialPathological tissue biopsies were collected from both in-patient and out-patient departments in Guangdong Province People's Hospital from December 2009 to May 2010.32 patients were diagnosed as nasopharyngeal undifferentiated non-keratinizing carcinoma.18 patients were diagnosed as nasopharyngeal chronic inflammation.MethodReal-time PCR and western blot were used to detect the Cyr61 mRNA and protein expression levels respectively in the tissues of nasopharyngeal carcinoma and nasopharyngeal chronic inflammation patients. SPSS17.0 was used to analyze the relevance between Cyr61 expression level and different types of syndrome in TCM. Result①It was observed that there was no difference between the Cyr61 mRNA levels in the in the tissues of nasopharyngeal carcinoma and nasopharyngeal chronic inflammation patients by Real-time PCR (P>0.05).(2)34 cases of nasopharyngeal carcinoma group (91.2%) showed Cyr61 protein positive, while only 2 cases of nasopharyngeal chronic inflammation group (11.1%) were positive. This data indicated that Cyr61expressed much higher in the tissues of nasopharyngeal carcinoma patients (x 2=32.53, P=0.00<0.05). The expression of the two groups, NPC was 1.14±0.42, chronic inflammatory tissue was 0.36±0.21, There was no significant difference (t=7.3, P=0.003 <0.05)③34 cases of nasopharyngeal carcinoma, in 27 for men, and 7 cases of women. The 27 cases of nasopharyngeal carcinoma in men, Cyr61 protein expression in 25 cases, the positive rate was 92.6%,7 cases of female NPC in the Cyr61 protein expression in 6 cases, the positive rate was 85.7%, statistically no significant difference between them(x2=0.327, P=0.51>0.05). The expression level of male is 1.10±0.43, and female is 1.29±0.41, There was no significant difference (t=-1.04, P=0.94>0.05)④34 cases of newly diagnosed patients with nasopharyngeal carcinoma in 17 patients (50%) of blood stasis syndrome and 15 patients (44.1%) of phlegm syndrome; And 14 cases of blood stasis syndrome (82.4%) showed Cyr61 protein positive, all cases of phlegm syndrome (100%) showed Cyr61 protein positive, Cyr61 protein expression of these two syndromes was no significant difference. And The expression level of them was 1.19±0.50 and 1.08±0.33, there was no significant difference (t=0.71, P=0.08>0.05)ConclusionCyr61 protein is generally expressed in nasopharyngeal carcinoma tissues. And it may determine the clinical types, stages and even prognosis for nasopharyngeal carcinoma patients.
