Background and Aims:The bioactive agents such as sodium butyrate and la, 25-Dihydroxyvitamin D3 have a potential anti-tumor effect. The aim of the study was to investigate the effects and mechanisms of sodium butyrate and 1α, 25-Dihydroxyvitamin D3 on the growth of HT29 cells.Methods:HT29 cells were treated with sodium butyrate and la, 25-Dihydroxyvitamin D3 in different dose or manner. The proliferation activity of HT-29 cells was measured by MTT assay. Cell cycle and apoptosis were assessed by flow cytometry and hTERT mRNA was determined by RT-PCR.Results:Sodium butyrate(0.5 mmol/L-2.0mmol/L) or 1α,25-Dihydroxyvitamin D3(10"8mol/L-10-6 mol/L) alone inhibited the growth of HT29 cells in time- and dose-dependent manner. The rate of inhibition was more obvious in the group of sodium butyrate combined with 1α,25-Dihydroxyvitamin D3 than that in the group with sodium butyrate or 1α,25-Dihydroxyvitamin D3 alone (P<0.05).Sodium butyrate (1.0mmol/L) or 1α,25-Dihydroxyvitamin D3 (10-7mol/L) increased the proportion of Go/G1 cells, decreased the proportion of S phase cells, induced cell apoptosis and down-regulated the expression of hTERT mRNA (P<0.05). Compared with sodium butyrate (1.0mmol/L) or 1α,25-Dihydroxyvitamin D3(10-7mol/L) alone, co-administration of sodium butyrate (1.0mmol/L) and 1α,25-Dihydroxyvitamin D3 (10-7mol/L)was more obvious effects on suppressing the proliferation of HT-29 cells, arresting cell cycle and inducing apoptosis, concomitantly with a decrease of hTERT mRNA expression than that in the group with sodium butyrate or la, 25-Dihydroxyvitamin D3 alone (P<0.05).Conclusions:Sodium butyrate and 1α,25-Dihydroxyvitamin D3 can inhibit the proliferation of HT-29 cells. The combination of sodium butyrate and la, 25-Dihydroxyvitamin D3 can exert synergistically inhibitory effect on the growth of HT29 cells. The mechanism maybe relate to the effects of arresting cell cycle, inducing cell apoptosis and down-regulating the expression of hTERT mRNA on HT-29 cells.
