| Objective: To study the effects of TGF-β1 and sodium butyrate on human colorectal cancer cell line HCT-116 proliferation.Methods: Human colorectal cancer cell line HCT-116 were treated with different doses of TGF-β1 , sodium butyrate and both of them.The methylthiazolyl tetrazolium assay(MTT) method was used to detect cell proliferation and draw a curve.The cell cycle was determined with flow cytometry.Results:1.TGF-β1 could significantly inhibit colorectal cancer HCT-116 cell proliferation;within the same time of incubation,this inhibition effect increased with increasing doses of TGF-β1; at the dose of 10ng/ml and 20ng/ml ,the inhibition effect increased with time.At the dose of 5ng/ml, no statistical difference of the inhibition effect was seen wtihin the incubation time of 48 hours and 24 hours (P > 0.05); but the inhibition effect in 72 hours was significantly higher than in 24 hours and 48 hours (P < 0.05).2. Sodium butyrate can significantly inhibit colorectal cancer HCT-116 cell proliferation; within the same time of incubation,this inhibition effect increased with increasing doses of sodium butyrate; at the dose of 0.5mmol/L,1mmol/L and 2 mmol/L ,the inhibition effect increased with time(P < 0.05).3. TGF-β1 (5ng/ml) in combination with sodium butyrate (0.5mmol/L) can significantly inhibit colorectal cancer HCT-116 cell proliferation(P <0.05);the inhibition effect increased with time (P < 0.05). Within the time of 24,48 and 72 hours, the inhibition effect of TGF-β1 in combination with sodium butyrate was significantly higher than TGF-β1 and sodium butyrate alone .(P < 0.05).4. The incubation of TGF-β1 10ng/ml,sodium butyrate 1mmol/L,TGF-β1 5ng/ml in combination with sodium butyrate 0.5mmol/L,an obvious increase of Gl phase cells (P < 0.05)and a significant decrease in the ratio of S phase(P < 0.05) and G2 phase cells (P < 0.05)were detected in all groups.Conclusion:1.TGF-β1 could significantly inhibit colorectal cancer HCT-116 cell proliferation; within the same time of incubation, this inhibition effect increased with increasing doses of TGF-β1. TGF-β1 through cell cycle exert influence the growth of human colorectal cancer cell line HCT-116 inhibition.2.Sodium butyrate could significantly inhibit colorectal cancer HCT-116 cell proliferation; this effect was shown in a dose-dependent and time-dependent manner. Sodium butyrate through cell cycle exert influence the growth of human colorectal cancer cell line HCT-116 inhibition..3. The inhibition effect of TGF-β1 in combination with sodium butyrate to colorectal cancer HCT-116 cell proliferation was significantly higher than TGF-β1 and sodium butyrate alone. It could be seen that TGF-β1 and sodium butyrate could synergize to increase the inhibitory effects on the proliferation of human colorectal cancer cells. |
