Netilmicin, a semisynthetic aminoglycoside antibiotic, has been widely applied to the treatment of gram-negative microorganisms. It has unique position in similar aminoglycoside drugs because of its advantages such as with wide antibacterial spectrum, less nephrotoxic and ototoxic potential than other commonly used antibiotics, and adaptation to many symptoms. In this paper, three methods with simplicity, rapidness, and sensitivity have been established for the determination of netilmicin by resonance Rayleigh scattering using bromocresol green, bromophenol blue and silicon molybdate as probes.1. Study on the interaction between netilmicin and bromocresol green by resonance Rayleigh scattering and its analytical applicationThe scattering intensity of bromocresol green or netilmicin is very weak in pH 1.09 HCl-NaAc buffer. However, the interaction between netilmicin and bromocresol green will result in the enhancement of RRS and appearance a new RRS spectrum.The maximum RRS peak located at 606 nm. The scattering intensity was proportional to the concentration of netilmicin in a certain range. The detection limit was 30 ng/mL. The RRS spectral characteristics of the system, appropriate reaction conditions, and the effects of coexisting substances have been investigated. The experimental results showed that this method has good selectivity and can be used for the determination of netilmicin with satisfactory results. In addition, the interaction mechanism was discussed.2. Resonance Rayleigh scattering spectral characteristics of the bromophenol blue-netilmicin system and its analytical application In pH 0.91 HCl-NaAc buffer, a resonance Rayleigh scattering method was developed for the determination netilmicin based on the interaction between bromophenol blue and netilmicin. The spectra characteristics, the optimum reaction conditions and the influencing factors have been discussed. Under optimum conditions, there is a good linear relationship between the relative RRS intensity and the concentration of netilmicin in a certain range. The method has been applied to determination of netilmicin in synthetic samples with satisfying results.3. Resonance Rayleigh scattering method for the determination of netilmicin based on the interaction between silicon-molybdate and netilmicin In pH 4.78 BR buffer solution, silicon molybdate could react with netilmicin to form a complex, which can result in the great enhancement of resonance Rayleigh scattering intensity and appearance of a new RRS spectrum. The maximum wavelength of RRS was located at 384 nm. The relative RRS intensity was proportional to the concentration of netilmicin in a certain range. The spectra characteristics, the optimum reaction conditions and the influencing factors have been discussed. The method has been satisfactorily applied to determination of netilmicin in synthetic samples.
