| Diabetes which called XiaoKeBing in Chinese medicine has become the highest rates of disease after the tumor and the cardiovascular. At present, the oral medications to treat diabetes in clinical are mainly sulfonylurea, double metformin kind, alpha-grape indicant enzyme inhibitors and so on, but will have different extent of damage for liver and kidney when long-term use. The traditional Chinese medicine has the long-term medication history in our country, relies on its multi-ingredient multi-target of the therapeutic mechanism, small side effects, good drug resistance and so on superiority, has appeared great value for study and development potential. Trichosanthis Radix as XiaoKeBing prescription treatment of universal used in one of the traditional Chinese medicine, which high frequency of its effective ingredients treatment for diabetes research but rare reports in the literature. Therefore, further study and confirmation the hypoglycemic active ingredients of Trichosanthis Radix has very important practical significance for hypoglycemic drug development.The main content of this study:(1) Screening the glucose-lowering activity of trichosanthis radix and studying its pharmacological effects.We extracted Trichosanthis radix with three methods:ultrasound with ethanol, reflux with ethanol, ultrasonic with water, then investigated the hypoglycemic activity of them through the alloxan-induced diabetic mice model. Using the most effect extraction, we further isolated the active components by macroporous resin D101 and silica gel column chromatography. The result was validated by calculating and comparing the consumption of the glucose in the cell model.(2) The improvement effect of Trichosanthis radix for diabetes complication.Through feeds diabetes mouse for the water elution component (groupâ… ), 20%-60% elution components (groupâ…¡),80%-95% elution components (groupâ…¢), determining the MDA content, the SOD level and the kidney/body weight index of the mouse blood serum finally.The results were as follows:Experiment (1) results show that:the Trichosanthis radix was extracted with ethanol in ultrasonic sound has the significant hypoglycemic effect and improve the symptom of diabetes mouse. The fraction of 80% elution is better on the glucose consumption and has shown obvious difference between the model group and other elution of groups (P< 0.001).Then we isolated two compounds and two mixtures from the 80% fraction, among then which mixture I shown obvious difference between the model group and other elution of groups (P< 0.05).therefore The fraction of 80% elution is one of the hypoglycemic activity constituent and mixture I maybe the hypoglycemic active component.Experiment (2) results show that:Trichosanthis radix can enhance diabetes mouse organism SOD activeness, reduces the MDA content, and has certain restore function for the kidney of diabetes mouse. The level of MDA and SOD of 80%-95% elution groups (group III) has significant difference between the model group (P< 0.01). The result was consistent with the experiment (1) of cell model measures and further confirmed ultrasound with ethanol of 80% elution is one of the hypoglycemic activity constituent. |
