Objective: This subject is based on the previous research of the research group in Gansu Province,the three main producing areas of different months of Radix Hedysari as the object of study,and provide a theoretical reference for the timely harvest of Radix Hedysari.The contents of four kinds of isoflavones in different habitats and different months were studied.The contents of polysaccharides and total flavonoids in different habitats were compared,and the theoretical basis was provided for the control of the quality of Radix Hedysari.The changes of four isoflavones and their ratios in different months were analyzed,which provided some reference for the intrinsic relationship between Rhizome and Active ingredients.Methods: The determination of the relevant components of the astragalus samples was carried out according to the moisture determination method(drying method),the ash method and the leaching method(hot-dip method-alcohol-soluble extract)in the first part of the Chinese Pharmacopoeia 2015.The content of total polysaccharides and total flavonoids in the astragalus samples was determined using ultraviolet spectrophotometry.The simultaneous determination of 4 isoflavones in samples by high performance liquid chromatography.HPLC was used to determine the content of four isoflavones in Radix Hedysari from different areas in Gansu Province,and the proportion of its components was calculated.Results: The length,diameter and weight of the samples increased with the increase of the growing months.The moisture content of the Radix Hedysari samples from Wudu Anhua in different collection months ranged from 5.16% to 6.41%,the total ash content ranged from 3.46% to 5.65%,and the content of Alcohol soluble extract was about 27.68% to 34.01%;The moisture content of the Radix Hedysari samples from Tanchang in different collection months ranged from 4.90% to 5.51%,the total ash content ranged from 3.46% to 5.65%,and the content of Alcohol soluble extract was about 24.72% to 58.94%;The moisture content of the Radix Hedysari samples from Longxi Shouyang in different collection months ranged from 4.63% to5.41%,the total ash content ranged from 4.82% to 6.25%,and the content of Alcoholsoluble extract was about 22.78% to 35.70%.In addition to the total ash content and the content of the extracts in the 4 samples do not meet the requirements of the Pharmacopoeia,the results of the other items are consistent with the provisions of the2015 edition of the Chinese Pharmacopoeia.The content of different origin samples in Radix Hedysari polysaccharide and total flavonoids were significantly different,And there was a negative correlation between the two;The content of total polysaccharide in Wudu Anhua Town,the highest content of total flavonoids in Tanchang Jiangtai;The samples in different months of Radix Hedysari polysaccharide were highest in October,the total flavonoids content was highest in July.There were significant differences in the content of four isoflavones(P<0.01),and there were significant and minimum values.In the three places of Radix Hedysari,the contents of isoflavones,isoflavones glucoside,formononetin and mannacles were as follows: July,August,September and October were the samples of Longxi Tanchang> Wudu samples,The three content of isoflavones and isoflavones glucoside and mangonite in Radix Hedysari were the lowest in October,and decreased from September to October.The content of Miscanthus sinensis was increased from September to October.In 7,8,9,10 months,Wudu,Tanchang,Longxi,Radix Hedysari characteristic components of the sample as calycosin glucoside.Cluster analysis showed that the samples collected in different months could be clustered into two groups.According to the different areas of the 7,8,9,10 months to match the chromatogram display,Samples from Wudu Anhua area has 25 common peaks;Tanchang area samples have 34 common peaks;Longxi Shouyang area has 25 common peaks.All the 12 samples were matched by chromatographic peaks.The results showed that there were 11 common peaks in each of the 12 samples.Superposition of peak area of common peaks in three major areas was Tanchang highest in October.Conclusions:There are some differences in the content of traits,moisture,total ash,alcohol extract,polysaccharide and total flavonoids in different regions in the sample in medicinal Radix Hedysari,and this difference is related to different origin.It seems that the quality of Radix Hedysari from Longnan Tanchang area is better than wudu Anhua and Longxi Songyang.Combined with the content and yield of eachcomponent,the best sampling time is 10 months late,and this conclusion is consistent with the traditional harvest time of Radix Hedysari.Accumulation of comprehensive analysis in different regions in the active components of Radix Hedysari also dynamic should be combined in different regions and in various ecological factors(light,precipitation,the annual amount of radiation,heat,etc.)influence the comprehensive function and soil nutrients on the contents of active components in Radix Hedysari.
