Study On Seroprevalence Of HP0596 In Chronic Gastritis And Gastric Cancer

Background and Objective:A product of the Helicobacter pylori Hp0596 gene(Tip-α) is a highly immunogenic homodimeric protein,unique for this bacterium. Tip-αis functionally unrelated to VacA, CagA and Urease.Tip-αis unique in H.polyri:No similar genes have been identified in sequence in the public database,except HP-MP1 of H.polyri strain SR7791 and jhp0543 of strain J99.In addition, Tip-αprotein has been found in clinical isolates of H.polyri. A case-control study comparing their HP0596 seroprevalence of antibodies with Gastric cancer (GC) and chronic gastritis(CG) and soldiers was carried out using enzyme -linked immunosorbent assay (ELISA) to analyze HP0596 relationship with chronic gastritis(CG) and Gastric cancer (GC) and normal people.Materials and Methods:Study subjects were histologically diagnosed 333 gastric cancer patients and 241 gastritis patients who visited Chinese PLA General Hospital or 261 Hospital from July 2009 to May 2010 and 235 normal people.Histological examination,H.pylori culture,Rapid urease test,Serum anti-H. pylori SS1 antibody ELISA and urease colloidal gold test were used to evaluate H. pylori infection. In order to identify whether HP0596 correlates with clinical outcome, a case-control study comparing their seroprevalence of antibodies with Gastric cancer (GC) and chronic gastritis(CG) and normal people was carried out using enzyme -linked immunosorbent assay (ELISA).Results:The H.pylori infection rate is 71.77% in gastric carcinoma patients compared to 82.99% in gastritis patients to 62.55% in normal people, there is significant difference among these three groups in infection rate (P<0.05), There are statistical difference in the rate of H.pylori infection among Gastric cancer (GC) and chronic gastritis(CG) and normal people. The HP0596 seroprevalence among gastric cancer and chronic gastritis(CG) and normal people were 29.73%%, 38.59%,25.11% respectively.The HP0596 seroprevalence of antibodies in gastritis patients is higher than them in gastric carcinoma patients and normal people. there is significant difference among gastric cancer and chronic gastritis(CG) and normal people in HP0596 seroprevalence (P<0.05). there is no significant difference between gastric carcinoma patients and normal people in HP0596 seroprevalence(P>0.05). Gastritis patients:OR=1.87, gastric carcinoma patients:OR=1.26, Gastritis patients is 1.48 higher than gastric carcinoma patients.Conclusions:1,The H.pylori infection rate of Gastritis patients is highest,and gastric carcinoma patients is higher,and normal people is low.2,This studies suggested that person infected H.pylori with Hp0596 have higher risks of gastric inflammation.3. This studies suggested that person infected H.pylori with Hp0596 have no relationship with gastric cancer.Acording to the seroprevalence of antibodies with chronic gastritis(CG),it maybe play a important role in gastric cancer,when it is Gastric cancer (GC), Hp0596 do less.
