Objectives:By studying the expression of BRCA1 and P53 protein on ovarian epithelium cancer,benign epithelial ovarian tumors and normal ovarian tissue, the relation between expression degree and histologic type of ovarian epithelial cancer,surgical pathology staging,pathological grading,lymph node metastasis respectively and their correlation and its roles played in the ovarian epithelial cancer,this paper provide us with certain theoretical reference for treatment and assessment of ovarian cancer.Methods:The fresh tissue samples resected from operation from January,2008 to October,2010 in CHINA-JAPAN UNION HOSPITAL OF JILIN UNIVERSITY and Changchun Central Hospital are fixed in 10% neutral formalin and embedded in paraffin with 4μm section thickness.Each sample was prepared with three slices,conducted with hematoxylin-eosin staining,immunohistochemistry staining and P53 immunohistochemical staining and the known positive slices was taken as positive control with PBS being replaced by one resistance negative blank control.All slices are blindly affirmed by pathology expert and accord with pathological diagnosis.Immunohistochemistry technique will be applied to stain tissue and to examine the expression of BRCA1 and P53 protein.Results:Expression products of BRCA1 protein are located in cell nucleus or cytolymph.Positive expression rate of BRCA1(30.8%) in ovarian epithelium cancer tissues is clearly lower than that of normal ovarian tissue(100%) and benign epithelium ovarian tumor tissue(86.7%) with remarkable difference(χ2 =32.14,p<0.05),while the expression of BRCA1 protein in benign epithelial ovarian tumors continue the downward trend when compared with its expression in normal ovarian tissue and benign epithelial ovarian tumors.Expression products of P53 protein are located in cell nucleus.Positive expression rate of P53(70.8%) in ovarian epithelium cancer tissues is higher than that of normal ovarian tissue(0%) and benign epithelium ovarian tumor (6.7%) with remarkable difference(χ2=37.34,p<0.05)while the expression of P53 protein in epithelial oarian cancer tissue continue the upward trend when compared with its expression in normal ovarian tissue and benign epithelial ovarian tumors.Expression rates of BRCA1 and P53 of different histological type are, 45% serous cystadenocarcinoma,20% mucicous cystadenocarcinoma,40% endometrioid carcinoma;positive expression rate of P53 are 70% serous cystadenocarcinoma,66.7%mucicous cystadenocarcinoma, 30% endometriosis carcinoma and different histologic type epithelial oarian cancer BRCA1 positive expression rate of any significant comparative differences (χ2 =1.88,p>0.05),while there is little difference in the expression rate of P53(χ2=2.14,p>0.05);positive rate of different histological grade are 45.7%G1~G2 grade,13.3%G3 grade.There is great difference of BRCA1 in positive expression betweenG1~G2 and G3 grade(χ2 =7.95,p<0.05),positive rate of P53 are 48.6%G1~G2 grade, 90% G3 grade respectively with great difference in positive expression betweenG1~G2 and G3 grade(χ2=12.68,p<0.05);In operation-pathological staging, there are 40cases I~II stage and 25 cases III~IV stage with positive rate of BRCA1 in I~II stage and III~IV stage respectively being 30% and 28%.There is little difference between them(χ2=0.03,p>0.05).Positive rate of P53 in I~II and III~IV grade are separately 75% and 64% with little difference (χ2=0.90,p>0.05);In ovarian epithelium cancer,there is no lymph node metastasis and the positive rate of BRCA1 is 75%,while that is 8% in ovarian epithelium cancer with lymph node metastasis;There is difference between(χ2=27.63,p<0.05);positive rate of P53 in ovarian epithelium cancer with no lymph node metastasis is 75%,while that in ovarian epithelium cancer with lymph node metastasis is 68%;The difference are distinct(χ2=0.38,p>0.05).There 65 cases positive expression of BRCA1 protein in ovarian epithelial cancer and 16 cases negative expression of P53 protein.Negative expression of BRCA1 protein and positive expression of P53 protein are 30 cases.There are 15 cases positive expression and 4 cases negative expression for both.According to it,we can find that there will be more shortage of BRCA1 protein expression and more expression of P53 protein in ovarian epithelial cancer which show that there exist mutual coordination between BRCA1 and P53 in the development of ovarian epithelial cancer.Spearman rank correlation analysis show that there exist negative correlation between them with r= -0.431,p<0.001.Conclusion:1.When compared with the positive expression rate of BRCA1 protein in epithelial oarian cancer tissue,that in normal ovarian tissue and benign epithelial ovarian tumors continue the downward trend which show that low expression of BRCA1 protein may promote occurrence and development of epithelial ovarian tumors.2.When compared with that in normal ovarian tissue and benign epithelial ovarian tumors continue the upward trend,the positive expression rate of P53 protein in epithelial oarian cancer tissue which show that high expression of P53 protein many promote occurrence and development of ovarian epithelial cancer.3. Low expression of BRCA1 protein is relevant to pathological grading and grade malignancy.The more high grade malignancy and pathological grading,the worse patients' prognosis which show that BRCA1 is likely to be the indicator predicting the prognosis of ovarian cancer.4.The high expression of P53 protein in ovarian epithelial cancer is related to pathological grading because the higher pathological grading,the worse patients' prognosis which show that P53 is likely to become the indicator which can predict the prognosis of ovarian epithelial cancer.5.There exist negative correlation between the expression of BRCA1 and P53 protein in ovarian epithelial cancer.To combine them together and examine jointly probably increase the diagnosis rate of ovarian epithelial cancer which further help us understand grade malignancy of ovarian cancer.It is the better auxiliary diagnosis methods and is expected to become the indicator to decide the prognosis of ovarian epithelial cancer...
