Objective: By observing the protective effect of Zhilong Huoxue Tongyu Capsule (ZHTC)on learning and memory function and hippocampus neurons apoptosis ,from the perspective of apoptosis ,to assess the curative effect of ZHTC and investigate its possible mechanism Methods : Vascular dementia model was duplicated by means of permanent occlusion of bilateral common catery(CCA),then they were divided randomly into the model group ,2g/kg piracetam group ,1.6g/kg,0.8g/kg and 0.4g/kg ZHTC group .sham operation group was another setted,each group was 10 rats.Then learning and memory ability of rats was tested with the Morris water maze.Stained with HE,the histological changes of hippocampal CA1 was observed.The MDA content and SOD and GSH-Px activity were determined with chemical-colorimetric method;The method of immunohistochemistry(SP)was used to detect the positive expression of Bax,Bcl-2 and Caspase-3 of hippocampal CA1. Adopt TUNEL method to detect the neurons in the hippocampus CA1 apoptosis. Adopt RT-PCR method to observe Caspase-3mRNA Positive express of the hippocampus histological. Results: Morris water maze results showed that the rats of every given medicine group were significantly lower than the average latency of the model group (P <0.05); Compared with sham operation group ,the content of MDA was obviously inereased,while the activity of SOD and GSH-Px was significantly decreased(P<0.05).HE staining results showed that the injury of hippocampal CA1 neurons of the rats every given medicine group was significantly ligther the model group,ZHTC can significantly improve the neuronal damage. TUNEL results showed that the positive expression MOD of rats in every given medicine group obviously was lower than model group (P < 0.05); Results of immunohistochemistry showed MOD of Bax and caspase-3 positive expression of treatment group was significantly lower than the sham operation group (P <0.05), while the Bcl-2 was significantly higher than the sham operation group. The result of Caspase-3mRNA was prompted with the same correlation with immunohistochemistry result. Conclusion ZHTC can improve learning and memory of VD rats. and were dose dependent ,can be used for the treatment of vascular dementia. The therapeutic mechanism may be as follows :(1)it can enhance SOD, GSH-PX activity and decreased MDA enzyme synthesis, thereby reducing oxidative stress caused by direct damage to hippocampal neurons. (2) increase the expression of Bcl-2 and reduce the formation of Bax to reduce the activation of caspase-3 to inhibit neuronal apoptosis.
