The creation of QutanHuoxue Decoction was guided with the theory of traditional Chinese medicine and combined with the achievement of modern pharmacology.During long term of clinical practice,Professor TongjiaoSun detected that QutanHuoxue Decoction had beneficial effect on fatty liver, and it is the research results of Eleventh Five-Year' National Technology Support Program on "clinical experience and academic research of Professor TongjiaoSun". In order to expound its pharmacological action and probe into its mechanism of action,the task had observed the prevention and cure function for QutanHuoxue Decoction in the treatment of non-alcoholic fatty liver.It will provide scientific basis for clinic and lay a foundation for the further research and exploitation of new drug.Methods:60 SD male rats were taken, under experimental conditions 1 week after feeding, were randomly divided into 2 groups:Normal control group (10), given normal diet; model group (50), given high fat diet. the model group Given high-fat diet after the second week,50 rats were divided into model control group, low dose treatment group, middle dose treatment group, high dose treatment group and Kaixilai drug treatment group. Low, medium and high dose group were given traditional Chinese medicine compound QutanHuoxue Decoction 0.5 times dosage (6g/kg.d),1 time dosage (12g/kg.d),1.5 times dosage (16g/kg.d). Kaixilai drug treatment group received Kaixilai drug 40mg/kg.d. Model control group,normal control group without any drugs, replaced the same dose of isotonic Na chloride. SD rats were free to drink water and eat, weighted 1 time every 2 weeks and we adjusted drug dosage accordingly. According to above drug plan,at the end of 8th,9th,10th weekend, We take 1-2 rats from the model group and killed them, Observe pathological changes in hepar-tissue and blood biochemical parameters (ALT,AST,TG,TC and so on). All SD rats killed at the end of 10th weekend, we measure hepatic function,blood lipid and observe result of hematoxylin and eosin stain,apoptosis in hepar tissue. Result: Model control group compared with the normal control group, its body weight, hepar weight and content of serum ALT, AST, TC, TG, range of hepatic pathological inflammation, degree of hepatic steatosis, index in hepatic apoptosis showed significant high expression (P<0.01); low dose treatment group, middle dose treatment group, high dose treatment group, Kaixilai treatment group compared with model group, their index are significantly lower or decreased (P<0.05);low dose treatment group compared with the model in body weight, the difference was not statistically significant (P>0.05); there is no significant difference in body weight between high and middle dose treatment group(P>0.05);Kaixilai treatment group compared with low and middle dose treatment group,their hepar weight demonstrate no significant difference (P>0.05);there is no significant difference in hepar weight between low and middle dose treatment group(P>0.05);compared with Kaixilai treatment group,their index on ALT,AST demonstrate no significant difference in low,middle and high dose treatment group(P>0.05); QutanHuoxue Decoction low-dose group compared with the control group, the difference between serum TG, TC was not statistically significant (P>0.05); there is no significant difference in range of hepatic pathological inflammation between high dose treatment group and Kaixilai treatment group(P>0.05); high dose treatment group, compared with model group, their index in degree of hepatic steatosis are significantly lower or decreased (P<0.05);there is no significant difference in degree of hepatic steatosis between high dose treatment group and Kaixilai dose treatment group(P>0.05);there is no significant difference in index on hepatic apoptosis between high dose treatment group and Kaixilai treatment group(P>0.05); Conclusion:QutanHuoxue Decoction can significantly reduce NAFL rat liver steatosis, re--gulate lipid metabolism disorders, to some extent, improve liver function, decrease the apoptosis index in liver,a control NAFLD, which may be th--e control mechanism of NAFLD.
