Objective: In order to explore the effects of Jiangzhiligan Decoction(JZLGT)on intestinal mucosal barrier of rats with non-alcoholic fatty liver disease(NAFLD).Methods: Forty male Sprague-Dawley rats were divided into normal chow diet(ND)group(10 rats)and modeling group(30 rats).NAFLD was successfully induced by high-fat diet(HFD)feeding for 12 weeks.Then the remaining 30 rats in the modeling group were randomly divided into model group(HFD)and low-(JZLGT-Low)and high-(JZLGT-High)dose Jiangzhi Ligan decoction groups(treated with the crude drug at 4.6 and 9.2 g /kg,respectively).Sampling was made 6 weeks later.Hepatic steatosis and NLRP3 inflamasome related proteins NLRP3 and Caspase-1 were detected to determined the function of JZLGT on NAFLD.Furthermore,the architecture of intestinal mucosal barrier,the the expression levels of tight junctions(TJs)proteins Occludin and zonula occludens-1(ZO-1)and portal endotoxin levels were also examined to explore the possible mechanism of JZLGT on NAFLD.Results: 1.Compared with ND rats,HFD rats showed obvious hepatic steatosis and upregulated NLRP3 and Caspase-1 protein levels.JZLGT-High rats displayed decreased adipocyte diameters and NLRP3 and Caspase-1 levels,suggesting high-dose JZLGT could ameliorate HFD-induced NAFLD.2.Compared with ND rats,the structure of intestinal mucosal barrier were sparse and irregular with the length of the intestinal villus dramatically decreased in HFD rats.Besides,the expression levels of Occludin and ZO-1 in HFD rats were significantly lowered.Compared with the HFD rats,the structure of intestinal mucosal barrier of JZLGT-High rats were more neatly arranged and less fractured with the length of intestinal villus obviously improved,the levels of Occludin and ZO-1 remarkably rescued.These results indicated that high-dose JZLGT could improve intestinal mucosal barrier function.3.Compared with ND rats,the portal endotoxin levels were notably elevated in HFD rats.Compared with the HFD rats,the portal endotoxin levels significantly declined in both JZLGT-Low rats and JZLGT-High ratsConclusion: High-dose of JZLGT can ameliorated intestinal mucosal barrier function of NAFLD rats,thus.Mechanically,High-dose of JZLGT can improve intestinal mucosal barrier function of NAFLD rats by improving the structure of intestinal villus and rescuing the expression of TJs proteins Occludin and ZO-1 and decreasing the portal endotoxin levels.
