Experimental Study Of Compound Ganshao Patch On Polycistic Ovary Syndrome Model Rats

Objective:In this project, the pathological models of rat were made. In order to explore the mechanism of the therapeuetic effect of Compound Ganshao patch on Polycistic ovary rats.Then, the influence of sex hormone, glucose, hepatic transaminases in serum and morphologieal changes of ovary were observed.Methods:Polycystic ovary syndrome rat models were established by letrozole(1mg/kgï¹’d, 21d). Seventy SD female rats, aged 6 weeks, in regular estrous cycle, were used and randomly divided into five groups: normal group,model group, Diane-35 group, Compound Ganshao capsule group and Compound Ganshao patch group. After drug intervention , rats were anesthetized with an intraperitoneal injection of 6% Chloral Hydrate, The blood was collected from vena cava, centrifugal settling, the serum was taken. The level of Alanine Amino transferase(ALT) and Aspartate Aminotransferase(AST) were measured by the coupled rate of enzyme, glucose was measured by glucose Oxidase-peroxidase and sex hormone was tested by radioimmunoassay (RAI). Double ovaries of all the ratswere resected and were put into 10%formalin for 24 hours. And then, tissue sectioned and HE dying. Mor-phologic changes wereobserved by ophthalmoscopy.Result:1.PCOS rat models were established by letrozole. Twelve days later, those rats were out of regular estrous cycle. Though JiRuis dying, leucocyte and a few keratinized cells were seen in the vagina smear, which pointed out that rats were at diestrus. Twenty-one days later, the level of serum sex hormone was tested. The level of LH, FSH and T was higher than the normal control group, but the level of E2, P was lower, those pointed out the PCOS rat models were successfully established2.After drug intervenetion, compaired with PCOS group rats,The serum level of LH, FSH, T of the Compound Ganshao patch group rats was lower and the level of E2, P was higher(P<0.05), but compaired with the other groups, The serum level of sex hormone were not changed(P>0.05).3.The Compound Ganshao patch group rats were with ruddy ovarian surface and thinner white coating. Mature folliculars and six to eight layers ovarian follicular granulose cells were seen by ophthalmoscopy, but no polycystic ovary was seen.4.Compaired with PCOS group rats, the serum level of glucose is lower(P<0.05), but the level of hepatic transaminases was not changed(P>0.05). Compaired with normal group, the serum level of glucose and hepatic transaminases were not changed(P>0.05). But compaired with Diane-35 group and Compound Ganshao capsule group, the level of hepatic transaminases was lower (P<0.05).Conclusion:1.The experimental results indicate that it is a fairly ideal method to establish polycystic ovary rat models by letrozole.2.Compound Ganshao patch harmonizes central nervous system and local ovay and adjusts incretion system of PCOS models, so that the level of LH, FSH, T, glucose are lower and the ovay issue are changed, so as to stimulate follicular development and ovulation. This is maybe the mechanism of Compound Ganshao patch to stimulate ovulation.3. Compaired with PCOS group rats, the level of hepatic transaminases of the Compound Ganshao patch group rats was not changed. But compaired with Diane-35 group and Compound Ganshao capsule group, the level of hepatic transaminases was lower, which indicates that it is a advantage for the patch that the burden of liver was not increased.
