Objective To recovery T24/ADM cells and detect the drug resistance. Matrine on T24/ADM cytotoxicity observed to determine the concentration of non-toxic dose range.To observe the in-fluence of matrine on T24/ADM to detect cell inhibition rate and expression of P-gp effect.To study the reversal of matrine on the role of drug resistance T24/ADM cells.Methods Detected drug resistance on T24/ADM. The tutor group will be frozen in the T24 cells with T24/ADM cell recovery, successful resuscitation training add into ADM (concentration for 0.0μg/ml,0.5μg/ml,1.0μg/ml,2.0μg/ml,4.0μg/ml). Cells were observed under inverted microscope, morphological changes and growth, MTT detect cells to drug tolerance, Flow cytometry the expression of P-gp.MTT detect of matrine on T24/ADM of toxicity.The matrin was grouped according to differ-ent concentrations and added into the T24/ADM to measured their cytotoxicity and to identify non-toxic concentrations. Experimental group was added matrine (concentration for 10μg/ml, 20μg/ml,30μg/ml,40μg/ml,50μg/ml,60μg/ml), control group only added an equal volume of medium without drugs.The non-toxic concentrations of matrine was grouped according to different concentrations (concentration for 0.0μg/ml,10μg/ml,20μg/ml,30μg/ml,40μg/ml,50μg/ml), and added in-to the T24/ADM to measured in each group to detect cell inhibition rate and expression of P-gp and determine the best reverse the concentration.Results Detection T24/ADM drug resistance. Inverted microscope, T24/ADM T24 cells with the morphological characteristics change significantly. T24/ADM of ADM significantly resistant (IC50=7.37μg/ml), The P-gp expression of T24/ADM was significantly increased. Analysis of variance, the difference was statistically significant between the concentration (P<0.05).Determination of matrine on T24/ADM cells toxic. Dose-response curve can be determined by the non-cytotoxic dose of MAT (the growth rate>95%) were 50μg/mL, Therefore, the non-cytotoxic dose range of concentration as a reversal.Matrine reversal reversal test and determine the best concentration. MTT detect of cell inhibition rate resistant cells percentage. Obtained using single factor analysis of variance A (without matrine) and the B1 group (10μg/ml) and the B1 group (10μg/ml) and B2 groups (20μg/ml) between the two groups was not significant (P>0.05), the rest of the group differences were statistically significant (P<0.05). Resistant cells by flow cytometry the expression of P protein, obtained by single factor analysis of variance B1 group (10μg/ml) and B2 groups (20μg/ml), B2 group (20μg/ml) and the B3 group (30p,g/ml), B3 group (30μg/ml) and B4 groups (40μg/ml) between the two groups was not significant (P> 0.05), the rest of the group differences were statistically significant (P<0.05). And determine the concentration of matrine maximum reversal effect when 50μg/mL.Conclusion Matrine had the reversal effect on T24/ADM cell resistance.
