ObjectiveTo study the deficiency symptoms of nervous function,water content of brain,morphological changes, change of intracellular calcium concentration and the effect on mannitol and Wulingsan after cerebral ischemia in rats.Methods1. Sixty four SD rats, male/female, weighing 260±20 g,were used to make models of middle cerebral artery occlusion embolized by suture,which were randomly divided into Mannitol-treated group(n=16),Wuling san-treated group(n=16), Mannito & Wulingsan-treated group (n=16)and model group(n=16) after three hours of artery occlusion. Eight rats of every group were randomly executed after five days and ten days of infarct respectively. Another eight rats without cerebral artery occlusion served as control group.2.Nervous function in rats were scored and the percentage of water content,pathologic detection by microscope and the change of intracellular calcium concentration in brain were determined.Results1.Nervous function in conscious rats after middle cerebral artery occlusion had no significant differences among model group,Mannitol-treated group,Mannitol & Wulingsan-treated group and Wulingsan-treated group>0.05).There were significant difference between Mannitol-treated group (2.33±0.49) and model group (3.00±0.73) after five days since ischemia; There were significant differences between Mannitol&Wulingsan-treated group (1.53±0.64) and model group (2.31±0.80) after ten days from ischemia.2.There were significant differences among Mannitol & Wulingsan-treated group,Mannitol-treated group and Wulingsan-treated group which could alleviate brain edema in contrast with model group (P<0.05) and the effect of Mannitol & WuLing Powder-treated group was the strongest.The water content of brain tissue decreased significantly after five days from ischemia among Mannitol-treated group (83.51±0.17),Mannitol & Wulingsan-treated group (84.03±0.13) and Wulingsan-treated group (84.40±0.23) in contrast with model group (P<0.01).There were significant difference between Mannitol & Wulingsan-treated group and Wulingsan-treated group.The water content of brain tissue after ten days of ischemia decreased in contrast to that after five days. There was no significant difference between Mannitol-treated group (80.80±0.62) and Wulingsan-treated group (80.63±0.55) (P>0.05),but there was significant difference between Mannitol-treated group and Mannitol & Wulingsan-treated group (79.91±0.37) (P<0.01)As the same time, there was no significant difference between Mannitol-treated group and Wulingsan-treated group(P>0.05), but had significant difference in contrast with control group,model group and Mannitol & Wulingsan-treated group(P<0.01).Medicine and time may had mutual effect on water content of brain tissue (F=55.17,P<0.01)3.HE staining of brain tissue from coronal section showed that morphological structure and interstitial space in control group were normal; model group showed spaces around vessels and cells widened,partial neuron and spongiocyte was swoln after five days. The cellular diseases mainly included cellular swelling,neuron contraction and denaturalization and necrosis of nervous cell; Mannitol-treated group,Mannitol-treated group and Mannitol & Wulingsan-treated group and Wulingsan-treated group reduced the spaces and cellular swelling degree in some extent.4.Intracellular calcium concentration after ischemia improved more obviously than that of control group. Intracellular calcium concentration elevated higher after five days than that after ten days.Intracellular calcium concentration in the Mannitol-treated group (605.14±5.52),Mannitol & Wulingsan-treated group (613.21±6.34) and Wulingsan (633.20±5.58) were significantly decreased as compared with that in the model group (644.39±4.93) (P<0.01) after five days. There were significant differences between Mannitol-treated group and Wulingsan-treated group,but had no significant difference as compared with Wulingsan-treated group.Intracellular calcium concentration after ten days decreased as compared with that after five days,there wasn't significant difference in the concentration of intracellular calcium between Mannitol-treated group (421.24±4.17) and model group (424.10±8.42) (P>0.05),but there were significant difference when Mannitol-treated group was compared with Mannitol& Wulingsan-treated group(382.98±3.15) and Wulingsan-treated group(403.08±3.78)(P<0.01).At the same time, there was no significant difference between Mannitol-treated group and wulingsan(P>0.05),but when Mannitol-treated group was compared with control group,model group and Mannitol & Wulingsan-treated group,there were significant differences. Medicine and time may have reciprocal effect on the intracellular calcium concentration.Intracellular calcium concentration is correlated positively with water content in brain tissue. Conclusion1.Mannitol-treated group,Mannitol & Wulingsan-treated group and Wulingsan-treated group decrease the deficiency symptoms of nervous function and accelerate the recovery of nervous functions.2.Mannitol & Wulingsan can reduce water content of brain tissue in rats with brain edema after ischemia injury and calcium concentration, effectively preserve blood brian barrier and the degrees of brain edema to decrease the pathological changes of ischemical injury.3.Mannitol & Wulingsan has bettertherapeutic effects than Mannitol or Wulingsan alone.
