ObjectiveTo detect the expression of cytokines and specific transcription factor of Thl7 cell investigate the expression and significance of Th17 cell in patients with rheumatoid arthritis (RA).MethodsFirstly, all the RA patients selected into this study were divided into different disease activity groups according to Disease Activity Score. The mRNA and protein expression of transcription factors of Th17 cell was detected by Real Time PCR and Western Blotting. The expression of serum IL-17, IL-21 and IL-23 was de disease activity groups according to Disease Activity Score. The mRNA and protein detected by Enzyme-linked Immunosorbent Assay.Results1. The expression of mRNA and protein of ROR-yt, IL-17. IL-21 and IL-23 showed significant difference between the RA group and control group.2. The expression of mRNA of ROR-yt in RA with DAS28>5.1 was higher than RA with3.25.1 was higher than RA with 3.25.1 was higher than RA with 3.25.1.2. The high expression of protein of ROR-yt, IL-17, IL-21 and IL-23 was only found in RA patients with DAS28>3.2.3. The expression of ROR-yt, IL-17, IL-21 and IL-23 in RA patients with DAS28^ 3.2 was close to the control group.4. It showed that Th17 cells may be involved in the disease activity of RA, and its expression became normal after remission. The study provided an experimental basis for Th17 cells as indicators of disease activity.
