Expessions Of PPARγ In Limbs Of TAO Rats

Objective:1,To establish the TAO models in rats though the injection of laurate sodium2,To observe the changes of PPARy expressions in TAO rats and discuss the importance it plays in the pathogenesis of TAO.Methods:1,Modeling and groups:30 male Wistar rats were randomly devided into 6 groups:the sham operation group and the operation groups which including 4 weeks,2 weeks,1 week,48 hours and 24 hours.5 rats per group. The operation groups were given laurate sodium 0.2ml within the concentration of lOmg/ml through the injection into their left posterior limbs. The sham operation group were given 0.2ml normal saline into their left posterior limbs. Each right posterior limb was regarded as blank control. Take both sides of the femoral arteries,veins and the skeletal muscle when experimental time is over.2,Testing methods:The expression of PPARγprotein level was detected by imunohistochemistry techeniques. PPARγmRNA level was detected by RT-PCR.3,Statistical MethodsThe results were expressed in terms of x±s. Analysis of variance and LSD were used and the results were obtained by SPSS 13.0 software.Results:1,General observationPaleness and lower temperature immediately occur in the left posterior limb.Within time prolonged,Rats in the operation groups showed different levels of ischemia including swollen,blacken,muscle atrophy,necrosis and. dropping of the limbs. Some showed purulent exudate.2,Pathalogical observation2.1 HE staining showed that the cytoplasm is pink and the nuclear is blue.There is different levels of thrombosis in the vascular. Large amounts of inflammation cells infiltrated in the muscle. Neovascular can be seen around the femoral artery.2.2 Imunohistochemistry showed the intimal elastic lumina is unbroken. Endothelia cells and the muscle fiber were dyed.3,Expression changes of PPARy mRNA and proteinExpression of PPARy mRNA and protein can be found in normal vascular endothelia cells and in skeletal muscle. However, the expression of PPARy declined in TAO rats with time prolonged. Except the 24h group, the expression of PPARy mRNA and protein levels in group 48h,1w,2w and 4w are significant compared with the sham operation group and the control group (P<0.01). There is also significant difference among these five operation groups(P<0.01).No significant difference was found among the right posterior limbs.Conclusions:1,TAO models were successfully established by femoral puncture and the injection of luarate sodium into vascular. It's easy and cheap to operate.2,Normal vascular endothelia cells and skeletal muscle can express PPARγ. The expression of PPARy dropped in terms of mRNA and protein levels which can be detected in TAO rats. It maybe involve in the inflammation reaction in TAO pathogenesis.
