OBJECTIVE: To study the expression of RECK (revemion-inducing-cysteine- rich protein with kazal motifs) and MMP-9 (matrix metalloproteinase-9) in laryngeal squamous cell carcinoma (LSCC) and adjacent tissues and their clinical significance. METHODS:Using RT-PCR and citric-SP-immunochemical technique,the expression of RECK and MMP-9 were studied in different regions of larynx squamous cell carcinoma including 38 cases of laryngeal carcinoma and adjacent tissues which was 0.2,0.5,1.0cm away from cancer. Bind with clinical follow-up visit analysis the relationship between RECK and MMP-9 expression and clinical date.RESULTS:The expression of RECK increased in sequence of cancer tissue,at 0.2cm,0.5cm,1.0cm adjacent to carcinoma in expression intensity and rate,and there was difference between every two groups(P<0.001).The expression of MMP-9 degraded the expression intensit and rate in the sequence of cancer tissue ,at 0.2cm,0.5cm,1.0cm adjacent to carcinoma,and there was difference between every two groups(P<0.001). RECK and MMP-9 there were statistically significant different between cancer tissue and 0.5cm,1.0cm distant from the tumor;there were statistically significant tissues adjacent 0.2cm from tumor compared with tissues adjacent 0.5cm and 1.0cm from cancer (P<0.05) ,but there was no statistically significant compared tumor tissues with adjacent tissues 0.2cm from tumor and compared the tissues adjacent 0.5cm with 1.0cm away from cancer(P>0.05). The expression of RECK was negatively correlated with the expression of MMP-9(P<0.05). Expression of RECK and MMP-9 were correlated with the clinical stages, histopathology differentiation and lymph node metastasis in squamous cell carcinoma (P<0.05).CONCLUSION : RECK in LSCC showed negative expression and MMP-9 was positive expressed . he expression of RECK and MMP-9 was negatively correlated ; RECK and MMP-9 have correlation with the clinical stages, differentiation degree and lymphatic move case in LSCC ; two indexes have apparent change in 0.5cm distant from the tumor .
