Objective: To investigate the variation of T helper cell cytokines,IL-4 and IFN–γin serum and amniotic fluid induced preeclampsia and normal pregnancy, and so to discuss the role of Th1/Th2 immune cytokines in pathogenesis of preeclampsia.Methods: 15 cases with light preeclampsia ,25 cases with severe preeclampsia and 20 cases of women with normotensive late pregnancy (NLP) were observed. ELISA was used to measure the levels of IFN-γand IL-4 in serum and amniotic fluid of three groups. Results: (1)In serum , the levels of IFN-γin light preeclampsia group(115.39±15.77 )and severe preeclampsia group(128.56±32.14) was significantly higher than normal pregnancy group(95.04±18.13)(P<0.05), The levels of IFN-γin light preeclampsia group and severe preeclampsia group was no statistical significance(P> 0.05);(2)In serum ,The levels of IL-4 in normal pregnancy group(41.92±4.13), light preeclampsia group(38.61±1.78) and severe preeclampsia group(36.40±6.46) was no statistical significance (P> 0.05);(3)In amniotic fluid ,The levels of IFN-γin severe preeclampsia group(122.93±60.20) was significantly higher than those of normal pregnancy group(66.02±23.47)and light preeclampsia group(92.51±12.13)(P<0.05),The levels of IFN-γin normal pregnancy group and light preeclampsia group was no statistical significance(P> 0.05); (4)In amniotic fluid,The levels of IL-4 in normal pregnancy group(14.83±0.98), light preeclampsia group(13.92±1.66) and severe preeclampsia group(13.83±0.81) was no statistical significance (P> 0.05)(;5)In serum ,The ratio of IFN-γ/ IL-4 in normal pregnancy group(2.27±0.60),light preeclampsia group(2.99±0.42) and severe preeclampsia group(3.68±1.06) was statistical significance(P<0.05);(6) In amniotic fluid ,The ratio of IFN-γ/ IL-4 in normal pregnancy group(4.49±1.69,),light preeclampsia group(6.71±0.95) and severe preeclampsia group(8.93±4.42) was statistical significance(P<0.05).Conclusion : The imbalance of Th1 and Th2 cytokines in serum and amniotic fluid of severe preeclampsia is one of the pathogenesis of preelampsia. It is closely related with the disease.
