Glycocholic acid (GCA) is one of the main active component of bile from cattle and sheep. The aim of the study is to investigate effects of GCA on apoptosis of splenic lymphocyte both in vitro and in vivo in mice. The morphologic features of splenic lymphocytes were observed under inverted microscope, agarose gel eiectrophoresis was used for examining DNA fragment of spleen lymphocytes, flow cytometry was used for examining the percentage of apoptosis on spleen lymphocytes which Serine had evaginated, the method of spectrophotometer was used for examining the activity of redox reaction after administer GCA—content of GSH, and ELISA was used for examining activity of Caspase-9 and Caspase-3. The results In vitro show that spleen lymphocytes produce karyopyknosis and apoptotic bodies after adding GCA of different concentration to spleen lymphocytes, DNA fragmentation was caused remarkably(P<0.05), and induce lymphcell in spleen Serine had evaginated(P<0.01), apoptosis rate,on spleen lymphocytes and activity of Caspase-9 and Caspase-3 were improved remarkably(P<0.05), and also content of GSH was reduced obviously(P<0.05). Conclusion:GCA has promote influence on apoptosis of splenic lymphocytes both in vitro and in vivo in mice.
