Background:Nerve - immune response regulation of relations is gradually gained much attention, the sympathetic in spleen play a important role in autoimmune and central nervous system- immune response regulation, it is also closely related to physiological function of immune cells, directly or indirectly through its regulation of blood within the spleen hemodynamics and the expression of several cytokines. ERK was the first to be found in mitogen-activated protein kinase (MAPK), which is a kind of serine / threonine protein kinase, whose family has five sub-family, namely ERK1-ERK5, the expression of a wide range. They pass the mitogen signal, control a variety of important cell biological processes, including cell proliferation, differentiation and apoptosis, even in the behavioral response and cognitive aspects such as learning, memory, etc. ERK1, ERK2 two sub- are the most widely distributed family and the most in-depth functional studies. ERK1,ERK2 are also known as p44MAPK and p42MAPK, relative molecular mass of 44 kD and 42 kD, are very important members of MAPK family, and through the Ras / Raf / MEK / ERK pathway in achieving its function. ERK signaling cascade can be specifically mediated by a wide range of biological processes, its mechanism mainly through the feedback control signal, the interaction with scaffold proteins, the change of subcellular localization, in the cascade have different functions for each link a variety of components, cell non-phosphatase inhibitor and G protein regulation achieved. Thus, ERK expression in spleen lymphocytes to a certain extent reflects cell proliferation, immune response and other functions.Objective:To study in the rat spleen under no sympathetic innervation, spleen lymphocytes changes in ERK expression, and comparative analysis with the normal spleen, trying to offer basis of splenic nerve - immune response regulation mechanisms.Methods:20 adult male SD rats which are clean grade, were randomly divided into experimental group and control group. Resection experimental group 10 rats spleen nerve fibers, keeping the spleen removed after 1 week, 4% paraformaldehyde, paraffin, HE staining and ABC immunohistochemical staining.1) First detection of PGP9.5 expression, using ABC immunohistochemistry to determine whether the experimental rats denervated spleen requirements, excluding the expression, only whether or not to discriminate; 2) conventional HE staining spleen were observed general differences in general structure; 3) spleen lymphocytes and the expression levels of ERK expression, the same ABC immunohistochemical method to detect the expression of lymphocyte ERK, counting every 5 doman 100 cells in the perspective of positive cells, using image-pro plus image analysis system, the positive part of the average absorbance (mean optical density, MOD), to represent the positive parts of the protein.Results:HE staining can be seen through the slice, two clear structure of spleen structure, periarterial lymphatic sheath cell dense lumps, red pulp shows a large number of red blood cells, removing the spleen innervation in rat experimental group than in the control group in general no significant changes in spleen structure.ERK expression in spleen lymphocytes in both groups, normal control group, the expression of ERK-positive cells in the spleen accounted for 16.2% (16.20±3.05), mainly distributed in the PALS; the experimental group only 6.0% (6.0±1.63), the relative distribution uniform, but still mostly PALS. ERK expression in spleen of the experimental group was significantly lower than the control group, the difference was significant (P <0.01). Using image-pro plus image processing system results seen in the experimental group MOD values (0.41±0.03) lower than the control group (0.41±0.03), and the difference was significant (P <0.01), we can see that after the removal of nerve , spleen cells ERK amount of normal spleen, but also shows that removing the nerves in the spleen, its activity was significantly decreased lymphocyte proliferation, which to some extent, affect the immune function of spleen.Conclusion:Removal of sympathetic innervation in the spleen, the spleen lymphocytes expressed in the number of ERK significantly decreased expression in the nucleus, especially the proportion of reduced, indicating decreased cell proliferation; ERK expression in normal spleen lymphocytes, mainly located in the nucleus , indicating active cell proliferation.
