Objective:To investigate the effect of saponins extracted from Kangaisan on induction of apoptosis in human hepatic carcinoma Bel-7402 cells and its molecular mechanisms.Methods:The growth inhibition of human hepatic carcinoma Bel-7402 cells was measured by MTT assay. The effect of saponins extracted from Kangaisan on induction of apoptosis of Bel-7402 cells was studied by light microscopy and flow cytometry (FCM), respectively. Immunocytochemical method was performed to analyze the expression of Bcl-2 and Bax proteins after treated saponins extracted from Kangaisan, Western blot analysis was used to detect the expression of Bcl-2 and Bax protein after saponins extracted from Kangaisan treatment.Results:saponins extracted from Kangaisan could significantly inhibit the growth of Bel-7402 cells, in a dose-and time-dependent manner, while. When Bel-7402 cells were exposed to saponins extracted from Kangaisan at 44μg/ml for 24 h,48 h, the sub-G1 cells were increased, in a dose-and time-dependent manner. The expression of Bcl-2 protein was down-regulated, and expression of Bax protein was up-regulated, by immunocytochemical method after exposure to saponins extracted from Kangaisan (44μg/ml,24 h,48 h,72 h). Western Blot analysis showed that Bcl-2 expression were decreased, while Bax expression was increased, after exposure to saponins extracted from Kangaisan(44μg/ml,24 h,48 h,72 h).Conclusion:Saponins extracted from Kangaisan could significantly induce apoptosis of human hepatic cancer cell Bel-7402, the apoptosis of human hepatic cancer cell Bel-7402 caused by saponins extracted from Kangaisan may be associated with down-regulation of Bcl-2 and up-regulation of Bax.
