Objective:To study the efficiency and mechanism of Chansu Capsule with medicated serum in inducing the apoptosis of human hepatocellular carcinoma(HCC) BEL-7402 cells and expression of Bcl-2, Fas, Apo2. 7 in apoptotic HCC cells.Methods:20 New Zealand albino rabbits were randomly divided into two groups (group 1 and group 2), each group including 10 rabbits using serum pharmacologic method, group 1 were prepared medicated serum containing chansu capsule, group 2 were prepared blank serum. We chose different concentrations of medicated serum containing chansu capsule as experimental groups, which were respectively co-cultivated with human HCC BEL-7402 cells for 24 and 48 hours. The morphological change of apoptotic HCC cells was observed with light microscope; the percentage of dyeing cells were examined by trypan blue; the cell growth was examined by methyl -tetrazolium (MTT) assay; the percentage of apoptotic HCC cells, the change of DNA and Apo2. 7 protein were examined by flow cytomericassays; the expression of Bcl-2 and Fas protein was measured with immunocytochemistric staining; blank serum was taken as control group.Results:Some cells of experimental groups become smaller and rounder, with single floating growth; dyeing cells were not remarkably change; the cell growth was remarkably inhibited by the medicated serum contaning chansu capsule; Gi-. S phase of cell cycle decreased, G2/M phase of cell cycle increased, and the apoptotic peak was observed by flow cytomeric assay, the percentage of apoptotic HCC cells change with the time and the concentration of medicated serum; the expression of Fas and Apo2. 7 protein in human HCC BEL-74O2 cells treated with chansu capsule obviously increase -ed, however, the expression of Bcl-2 protein decreased. Experiment -al groups were significantly different from the control group (/><(). 05).Conclusion:The chansu capsule could induce apoptosis of HCC BEL-7402 cells, the percentage of apoptotic HCC cells is related to the time and concentration of medicated serum, with obvious time-response and dose-response relationship ; Chansu Capsule suppressed the growth of BEL-7402 cells by inducing apoptosis, which may be related to the change of the Fas> Bcl-2 protein and the cell cycle. It indicated that Chansu Cpsule probably become a potential medicament of anti-tumor.
