Polychlorinated biphenyls (PCBs) are a class of organic compounds with 1 to 10 chlorine atoms attached to biphenyl which is a molecule composed of two benzene rings each containing six carbon atoms, while polychlorinated biphenyls (PCBs) are been widely used in transformers, capacitors and paper and paints industry. Polychlorinated biphenyl (PCBs) has attracted a lot of attention because of the harmful to the immune system, reproductive system, nervous system and endocrine system, even endanger the liver and cause of cancer. Polychlorinated biphenyls (PCBs) are one of 12 kinds which principally controlled pollutants according to Stockholm Convention. Both groups of compounds are chemically very stable and resistant to environmental degradation, and have longer half-life. They could do harm to the human healthy through the enrichment of food chain and the detection of the level of PCBs in organism has aroused extensive concern in the international community.To establish a sensitive and reliable analytical method for determination of polychlorinated biphenyls (PCBs) in human plasma.PCBs in human plasma were extracted ultrasonically with 5% Na2SO4,acetonitrile and 1-propanol/water(85:15) and cleaned up by solid phase extraction(SPE) with a C18 cartridge and determinated by gas chromatography with ECD(election capture detector).In this thesis, the effect of column with silica/sulfuric, the ratio of desorption reagents and denaturant method on the recoveries of PCBs were studied. According to the injection temperature, initial temperature and flow rate of gas saver, the optimal GC method has been developed.The main experiment and analytical results of this thesis were summarized as following: The recoveries of PCBs were higher than others when the sample was not treated by the column with silica/sulfuric. The optimal ratio of desorption reagents were composed of 3mL hexane and 1mL dichloromethane/hexane (1:4). The calibration curves of PCBs were linear (R>0.9995) from 1 to 50 ng/mL. The recoveries of the PCBs for human plasma fortified at the 5,20 ng/ml levels were 86.50%-120.00%.The within-day precisions were 1.76%-8.13%. The limit of detection of PCBs were 0.04-0.08 ng/mL.
