Natural botanic feed additive has advantages of good pharmacologic actions, non-specificity antibacterial action, lower residual hazard, less drug resistance and abundant resources. Green and innoxious feed additive has become the trend all over the world instead of antibiotics. But there are still many problems in this field: The quality standard was lack of standardization and standardization; the immunological mechanism was unclear; preparation process was unadvanced, and so on. Therefore, the key thing in this field is to improve the feed additive quality standard with the method of modern analysis technology, and to illustrate the molecular mechanism.The objects of this study are 3 chief productions of Shanghai Zhaoxiang Biotechnology Co. Ltd., in order to set a good example in this trade. Firstly, salicin, ursolic acid and apigenin were isolated from these feed additives. The average content of procyanidins is 18.96% with the method of ferric ions as catalysis agent; the average contents of monosaccharide and total saccharide are 26.89% and 43.99% with the method of 3,5-dinitrosalicylic acid colorimetry. Moreover, establish the fingerprint of Procyanidins feed additives as a standard for qualitative analysis. With the method of HPLC analysis, the average content of salicin is 13.14 mg/g, the average contents of ursolic acid and apigenin are 40.11 mg/g and 1.00 mg/g in the feed additives. After that, refer to the standard of hygiene and active ingredient, the quality standards of the feed additives are established. And then, the HPLC fingerprint of procyanidins feed additive is established as an important quality standards.In addition, the effect of 3 feed additives on immunity are analyzed with the model of mice splenic lymphocyte proliferation and mice macrophage pinocytosis, the secretory action of IFN-y, TNF-a and IL-1 is determined by ELISA. The result indicates, the procyanidins feed additive has the most activation. The proliferation of T lymphocyte reaches 201.13±5.20%, and the A value of pinocytosis reaches 0.460±0.033 at concentration of 100μg/mL. Moreover, the procyanidins feed additive can promote the secretory action of IFN-y in splenic lymphocyte and the secretory action of TNF-a and IL-1 in macrophage.
