Ozone is a strong oxidant.Many biologic effects have been attibuted to ozone,including increased glycolysis,effects on red blood cells,effects on rheology,bactericidal,fungicide,virustatic,immunomodulating action and analgesic.However,being used as a potent disinfectant for almost a century,its usefulness in medicine remains controversial.The problem should be settled because many patients undergo some form of ozone therapy every year. Ozone has toxicity of destroying respiratory apparatus function and damaging heredity,however there is not serious complication reported. In fact,ozone,like any other drug,is not necessarily toxic when used properly. The critical is density. It is reported that the safe density is between 1 ug/ml and 40 ug/ml. It is also reprted that methaemoglobin is undetectable and haemolysis is no higher than 1.0% when ozone dose is up to 80 ug/ml in blood. Different tissue has different tolerance to ozone.This experiment is to study initially joints,muscle and spinal dura mater tolerance to different density ozone,with Zelanian albino rabbits as experiment animals,by observing normal tissue's acute and subacute reaction to different density ozone,according to present information about ozone therapy,and it can provide basic data for further research and experimental data for clinical therapy.Methods:Two rabbits are singled out to cut their cartilage of knees,articular capsule,musculus triceps surae,spinal cord and spinal dura mater as normal control from 44 Zelanian albino rabbits,and 24 rabbits randomly singled out are divided randomly into two big groups:50ug/ml and 80ug/ml,and each big group is divided separately to six small groups. Articular capsule,cartilage and musculus triceps surae are adopted separately at one day,three days,one week,two weeks,one week and two weeks after injected into with ozone,and rabbits in the fifth and sixth small groups are injected repeatly at the interval of three days.18 remnant rabbits are divided randomly into two big groups:50ug/ml and 80ug/ml,and each big group is seperately divided to 3 small groups.Spinal cord and spinal dura mater are cut out separately at one day,three days and one week after ozone is injected.All tissues are imbeded in paraffin and dyeed with HE,and we can observe different influence to the same issue of different concentration ozone and the change of tisuue at different time point.Results:Perichondrium and cartilage matrix in two experimental groups remain integrated and synovium changes little.Cartilage matrix changes gradually from red to blue,which in 50ug/ml ozone groups recovers fully during two weeks,which in 80ug/ml one doesn't improve completely after two weeks.There isn't obvious difference between cartilage in the second group and that in the fifth one;Myofibrosis apears and aggravates in the firs,seond,third and fifth groups with 80ug/ml ozone,and inflammatory reaction occures in the second,third and fifth groups,and muscular fiber is approximate normal in the fourth and sixth group.Muscular fiber is also approximate normal in the firs,second,third,fourth and sixth groups with 50ug/ml ozone.Inflammatory reaction is finded in the fifth group;Spinal dura mater becomes raritas and thinner with 50ug/ml ozone;80ug/ml ozone can break spinal dura mater fibre,and it becomes obviously thinner.Conclusion:Both of the two concentrations ozone can accelerate proteoglycan degradation.Recovery of cartilage in high concentration groups needs more time.There isn't obvious difference between duplicate and single injection into article; 80ug/ml ozone can obviously destroys muscular tissues,so it can't be used clinically.50ug/ml ozone has a potential danger to musculature,then we suggest that clinical ozone concentration should be below 50ug/ml; 80ug/ml ozone can obviously destroys endorachis,50ug/ml ozone is relatively safe. So we suggest that clinical ozone concentration injected into vertebral canal should be below 50ug/ml.
