Influence Of Jiaji Electroacupuncture On Spinal Substance P In Inflammatory Rats

Purpose: To observe the changes of the spinal cord substance P on inflammatory pain rats treated by JIAJI electroacupuncture. To unveil preliminarily JIAJI EA. resisting the mechanism of hyperalgesia.Methods: To make the monoarthritis(MA) rats model with complete Freund's adjuvant(CFA).60 male Wister rats were randomly divided into Control group,Model group,Routine EA group , JIAJI EA group,JIAJI share Routine EA group,12 each group .After making model rats 1days,10days,14days,we observed the hyperalgesic behaviour scores. Observe the changes of theSpinal cord substance P with enzyme linked immunosorbent assay and immunohistochemistryafter the treatment.The EA method: In all groups,select KWD-808Ⅱimpulse EA apparatus,offerMethodical implse,freqencies per2second 60Hz and 2Hz in turn,resist 1mA,last 25min,everyday one time.In Routine EA group,select the rats'left"Zusanli","Sanyinjiao"points,up and down connects.In JIAJI EA group, select the rats'bilateral"Jiaji"points of C3 and L4, up and down connects of two groups'leading wires.In JIAJI share Routine EA group,the method is above-mentioned two ways shared use.Results:1.In 4days,10days,14days,compared with Model group,PWL in three Treatment groups prolong obviously (P﹤0.01), JIAJI EA group and JIAJI share Routine EA group have no obvious difference(P=0.56,P=0.73,P=0.48).In 10days,14days,compared with Routine EA group, JIAJI EA group and JIAJI share Routine EA group prolong obviously (P﹤0.05).2. Compared with Control group,the contents of the spinal cordsubstance P on inflammatory pain rats in Model groups rise obviously (P﹤0.01); compared with Model group,the contents of the substance P inall of EA groups reduce obviously (P﹤0.01); compared with Routine EA group, JIAJI EA group and JIAJI share Routine EA group reduce obviously (P﹤0.05); the contents of the substance P in JIAJI EA group and JIAJI share Routine EA group have no obvious difference(P﹥0.05).3. Compared with Control group,integrated optical density of positive cells of the spinal cord posterior horn substance P rise obviously (P﹤0.01); Compared with Model group, integrated optical density of the substance P in all of EA groups reduce obviously (P﹤0.01);compared with Routine EA group, JIAJI EA group and JIAJI share Routine EA group reduce obviously (P﹤0.05); JIAJI EA group and JIAJI share Routine EA group have no obvious difference(P﹥0.05).Conclusion: EA has obvious antagonist phentolamine on inflammatory pain rats,EA JIAJI is better than EA"Zusanli","Sanyinjiao"points.Itsanalgesic mechanism is likely to resist the convey of the spinal cordsubstance P on inflammatory pain rats.
