Objective: The main aim of this study was to investigate the change about production of Abeta40/42 and the expression of APP,BACE and PPARγin HEK293 cells stably transfected with APP695 containing the Swedish mutation,and the relationship among these changes,study the effects and mechanisms of rositazone on Alzheimer's disease,and provide the theoretical basis for treatment of Alzheimer's disease.Methods: In this study, HEK293 cells stably transfected with APP695 containing the Swedish mutation were given Rosiglitazone.We assayed the effect of Rosiglitazone on the proliferation and cytoxicity by MTT and lactate dehydrogenase(LDH) activity respectively ,the production of Abeta42 by ELISA,the expression of amyliod precursor protein,beta-secretase(BACE) and Peroxisome Proliferator-Activated Receptor.Results: We found that Rosiglitazone could significantly inhibit the expression of BACE,activate Proliferator-Activated Receptor and decrease the production of Aβ42 in dose-dependent and time-dependent manner in HEK293 cells,but could not affect the expression of APP.Conclusion: Rosiglitazone might decrease the production of A42 by Inhibiting the expression of BACE and activate Proliferator-Activated Receptor.
