Objective: To study the immune effect of the recombinant plasmid carrying IL-2 gene (pcDNA3.1/IL-2)on Hepatitis B virus vaccine(HBsAg and DNA vaccine). Methods: Using molecular cloning techniques, We constructed the recombinant plasmid carrying IL-2 gene ( pcDNA3.1/IL-2 ) , the plasmids(pcDNA3.1/IL-2,pcDNA3.1/S) were extracted large quantities by alkali lysis and purified by sepharose 4FF chromatography technique. Guinea pigs were devided into six groups: group 1 was control group; group 2 was HBsAg with Aluminum adjuvant; group 3 was HBsAg with IL-2; group 4 was HBsAg with plasmid pcDNA3.1/IL-2; group 5 was plasmids pcDNA3.1/IL-2 and pcDNA3.1/S; group 6 was plasmid pcDNA3.1/S. the plasmids(200μg) and HBsAg (10μg)were injected by intramuscular injection. one month later, to immune again. To isolate and detect the Guinea pig's sera every week in the first month and every two week a month later. we detected the anti-HBsAg antibody titer by ELISA and IL-2 activity by CTLL-2 cell line/MTT method. Results: We succeeded in constructing the recombinant plasmid pcDNA3.1/IL-2. After 12 weeks, the detected results of anti-HBsAg antibody titer were: group 2 was 2116mIU; group 3 was 1987mIU; group 4 was 2252mIU; group 5 was 67.19mIU; group 6 was 50.88mIU. IL-2 gene expression was observed that it increased in week one and declined after week one. The anti-HBsAg antibody titer and IL-2 activity in each group show significant difference(p<0.05). Conclusion: The recombinant plasmid carrying IL-2 gene (pcDNA3.1/IL-2)is a significant adjuvant to Hepatitis B virus vaccine.
