Background Adjuvants can quickly activate the immune system to enhance the immunogenicity of the vaccine.So far,there’ve been approved to apply to humans,including aluminum salts,MF59,AS03,AS04 and liposome,but there’s still some safety issues.Therefore,it is a core task to develop new high efficiency,economical and non-side-effect vaccine adjuvant.Glycerol-3-phosphate(G3P)is an important inducer of plant broad-spectrum immunization,which is similar to adjuvant and is the key to trigger immune response in plant.Evodiamine(EVO)is thought to be a promising anti-tumor drug in the future,and it can stimulate the immune system and promote the antigen-presenting effect of DC.Therefore,we hypothesized that G3P and EVO may have an adjuvant effect.Objective To investigate the effects of G3P and EVO on the humoral immune response in mice induced by hepatitis a vaccine.Methods The different doses of G3P and EVO were mixed with the hepatitis a vaccine as drug group,and the negative control group,the antigen group and the aluminum adjuvant group were set up;G3P 2mg,aluminum adjuvant and hepatitis a vaccine were mixed as compound adjuvant group,and negative control group,antigen group,aluminum adjuvant group,G3P group were set up.Each group of drugs immunized subcutaneously in ICR mice in once.The specific anti-HAV IgG levels in serum of mice were determined by enzyme-associated immunoassay(ELISA)in mice at 4,8,12 and 16 weeks after immunization.To observe the health status of mice in the experimental process,and at the end of the experiment,the viscera of the G3P 2 mg group,the EVO 80μg group mice,and the negative control group were observed for pathological sections.Results The anti-HAV IgG antibody was detected at 4,8,12 and 16 weeks after immunization except the negative control group.The antibody level of the G3P group was higher than the antibody level of the aluminum adjuvant group in the 4th week,and was statistically significant(P<0.05),and the G3P 2 mg group was the optimal dose group.The antibody levels of EVO 80μg group were higher than the antibody level of of the antigen group at 4,8,weeks after immunization,and were statistically significant(P<0.05).In addition,the experimental data show that the antibody level of G3P compound adjuvants were lower than the antibody level of the single adjuvants group,and compared with aluminium adjuvant group in 12 weeks after immunization,and were statistically significant(P<0.05);During the experimental process,the mice were in good health,and the pathological tissue sections were normal.Conclusions G3P and EVO have the effect of vaccine adjuvant,but the compound adjuvant group is not effective.
