Study on zinc supplement protection diabetic rats'kidney from oxidative tissue damageThe incidence of diabetes increases year by year, Diabetic nephropathy is one of the common serious chronic microvascula complication, and is the main cause of end-stage renal disease in chronic kidney disease,also had an important place in Chronic kidney disease.According to the American diabetes association,About 8℅percent of patients diagnosed diabetes at the same time had been diagnosed diabetie nephropathy. The pathogenesis of diabetic nephropathy is complex, in which oxidatie stress plays an important role.Zinc is a kind of indispensable trace elements in human body,also is a new type of antioxidants. Many studies show that diabetic patient had low serum zinc level . Diabetic nephropathy onset conceals, before occurring in clinical dominant proteinuria kidney pathological change yet, including increased glomerular hypertrophy and matrix which is important kind of clinical diabetic nephropathy pathological changes. Matrix metalloproteinases- 9 (matrix metalloprteinase 9, MMP-9) mainly involves extracellular matrix decomposition, Vascular endothelial growth factor (vascular endothelial have factor, VEGF) promotes vascular endothelial regeneration, Metallothionein is a strong kind of antioxidant. At present it is already clear that zinc can reduce diabetic retinopathy oxidative damage,and may induce myocardial cell to produce metallothionein ,which can mitigate diabetic myocardial cell damage. But complement zinc agents influence to diabetic kidney tissues are not clearly. This paper compare model group (STZ), external zinc intervention diabetic group (STZ+Zn) and external zinc intervention nomal group (NC +Zn) to nomal control group (NC), observe oxidation index content and renal MMP - 9, MT, VEGF expression. Further reveal Zinc supplements preparations preventiondiabetes mellitus resulting to kidney damage.provide a guidance, prevention and treatment methods to diabetic nephropathy . it has a good application prospect.Objective: To explore whether or not Zinc supplements could protect rat's kidney.Method: Wistar rats were randomly divided into four groups: Such as nomal control group (NC), model group (STZ), external zinc intervention diabetic group (STZ+Zn) and external zinc intervention nomal group (NC +Zn) . The diabetic models were made through intraperitoneal injection of streptozotocin(STZ) in STZ and STZ+Zn groups.The rats in STZ+Zn and NC +Zn groups were was via intragastric administration of 5mg·kg -1 Zn sulphate twice a week. After fed for 16 weeks ,the kidneys of Wistar rats were obtained and organ coefficient of kindey was calculated . measured the volume of superoxide dismutase(SOD) by the methods of xanthine oxidase, measured the volume of glutathione peroxdase (GSH-Px) by themethod of dithiobisnitrobenzoat , measured the volume of malonaldehyde(MDA) by the method of TAB, measured the volume of H2O2 measure the volume of H2O2 by the method of Molyadate,measured the volume of GST by the method of direct CDNB.The histopathological change of kindeys and the expressions of MMP-9,VEGF and MT were dected with immunohistochemistry. Data were presented as mean±SD. Statistical analysis was performed using SPSS 13.0 software..For enumeration data, a one-way ANOVA test were performed. For quantitative data, chi-square analysis was performed. P < 0.05 was considered to be statistically significant.Results.The volume of SOD and GST in the STZ+Zn group and the STZ group are higher than those of the NC group. The volume of GSH-Px in the STZ group are lower than that of the NC group.The volume of GSH-Px in the STZ group have rising tendentiousness compared with that of Zn group.Compared with normal control, the organ coefficients of in STZ group and STZ+Zn group was increased (P < 0.05). Compared with STZ group, the organ coefficients of STZ+Zn group was increased(P < 0.05). There was no difference between STZ group and NC +Zn group (P > 0.05).The MMP-9,VEGF and MT expressed in kidney of rats.Also MMP–9 and VEGF mainly expressed in the distal renal tubular,MT mainly expressed in the proximal tubularrenal tubular. There was no difference of expressions of MMP-9,VEGF and MT in kidney of various groups.Conclusion :There is oxidatie damage in diabetic rats′kidney.To a certain extent, zinc supplement maybe have a certain effect to oxidative damage of the kidney .
