Object:the resarch of microflora in gingival crevicular fluid around plants after operation is the mian part of resarch of oral cavity plant. DGGE is the electrophoresis technology to check DNA's mutation which is suggested by Fischer in 1979. it characteristic the microbial community by Nucleic acid, and it can more quickly check the nonculturable bacteria than traditional separation technology. PCR-DGGE is the technology which can check all kind of microbe in different kind of microbe. this research is based on every kind of people who meet experiment criterion, and check the preponderance flora's change procedure around plants gingiva in diffenent time after operation, discuss the oral Microbe influence to inflammation in gingiva in different time after plants operation. so it can provide clinic evidence. Methods:we select 20 patients who got treatment in chengdu Military Command organization hostipal, and their age is between 20-60. the plants' diameter is 3.75×8.5mm or 3.25×8.5 which is germany BEGO columnar, and we slect plants gingival crevicular fluid as specimen after 1 and 3 months when the operation is finished. and we slect health tooth's gingival crevicular fluid as reference specimen, pick-up these specimen's DNS and do operation analysis to compare the difference between plants and natural's gingival crevicular fluid. we use the SPSS 13.0 software to statistics.Results:1. brightness of DNA based on extract elisa in gene is higher than based on SDS cell Disintigrate. the time of extract DNA based on based on SDS cell Disintigrate is 2 times as based on extract elisa in gene. there are such addvantage such as DNA's concentration is higher, purity is good and save time.2. such as the result of 162 rDNA v3's PCR's display, every specimen display that every DNA, PCR's result is masculine.3 gingival crevicular fluid flora UPGMA comparability analysis:we can find by UPGMA comparability analysis,landscape orientation compare: every patient has small comparability between experimental group and reference group, and there is also conclusion that every patient has small comparability. portrait compare, two experiment has small comparability in experimental group, and there is also conclusion that every patient has small comparability.4. DGGE map's diversity analysis:there is comparability in biology richness of plants gingival crevicular fluid, natural tooth gingival crevicular fluid, gingival crevicular fluid of first moth and the gingival crevicular fluid of the third month.5. measure order result:plants group have no obvious difference with natural group, the main Microbes are:Uncultured peptostreptococcus sp.cloneN Fusobacterium simiae gene,Uncultured eukaryote clone,TM7 phylum sp.oral taxon 437 clone,Uncultured Porphyromonas clone and Uncultured Prevotellaceae bacterium,take capnocytophaga ochraceast as main flora to cultivate rare flora such as TM7 phylum sp.oral taxon 437 clone,Uncultured pasteurellales,Lautropia,capnocytophaga ochraceast; treponema and Fusobacterium simiae gene in gingival crevicular fluid belong to the third month and natural tooth are basic same. conclusion:the main flora is capnocytophaga ochraceast by PCR-DGGE to check superiority flora of plants and natural tooth in tow different time. the flora which can lead disease is easier to gather around plant in first month after operation. superiority flora is the basicly same in third month after the operation, and the rate of Anaerobacter is become higher. but there is no inflammation around plants. we can guide to Maintenance it as normal tooth to prevent inflammation.
