Objective To study the inhibition function of gene recombinant plasmid pDC316-hIL-24 withcisplatin on human cervical cancer nude mice model tumor growth and invasion.Method Nude mice, 4 weeks age,were injected with cervical cancer HeLa in2×107concentration into thire left central lateral subcutaneous. With construction of nude micemodel of human cervical cancer, the 30 nude mice were randomly divided into 5 groups, andrecombinant plasmids (groupⅠ) ,recombinant plasmids and DDP (groupⅡ)or DDP(groupⅢ),PBS (groupⅣ) as well as empty plasmids (groupⅤ) were injected into the tumor-bearing micewith Lipofectamine 2000 to interfere with the growth of transplanted tumor. Their effects on thevolume of the tumor and inhibitory rate of the mice were observed. hIL-24 expression wasdetermined in transplanted tumor by PCR. Lymph node metastasis was observed .Hematoxylin andeosin (HE) staining method was used to examine the tumor cell density, tumor cell mitoticcount ,and as well as immunohistochemistry was employed for investigating nm23H1 albumenin each group.Results①After transfection IL-24 gene was successfully transfected into and expressed intumor cells . Compared with the other two groups,ⅢⅣⅤtreatment group tumor volume wassmaller(P<0. 05),the average inhibition rate was higher(P<0. 05).The mice treated with IL-24combined with cisplatin had significantly smaller average tumor volume and significantly higheraverage inhibition rate (P <0. 05) than DDP only or IL-24 only group;butⅢⅣgroup have nosignificant different.②Pathological observation shows tumor cell mitotic count was decreased ingroupⅢⅣⅤ( P < 0.05).③Lymph node metastasis was decreased in groupⅢⅤ( P <0.05).Immunohistochemistry showed that, the expression of nm23H1 albumen in IL-24combined with cisplatin group and IL-24 group was significantly higher than other threegroups(P<0. 05).Conclusion: Gene recombinant plasmid pDC316-hIL-24 combined with cisplatin cansignificantly inhibit the growth of transplanted human cervical cancer in nude mice. And IL-24can strengthen the sensitivity of agreeable platinum chemotherapy medicine treatment, at thesame time , reduce the side effect of single medicine chemotherapy .IL-24 may activateexpression of nm23H1 albumen to inhibit human cervical cancer nude mice model tumor growthand invasion.
