Objective To explore the correlation of clinical general parameters and chemotherapyefficacy with the expression of P-glycoprotein(P-gp),TopoisomeraseⅡ(TopoⅡ) andGlutathione-s-transferases-π(GST-π) in untreated Non-Hodgkin's lymphoma.Methods (l)To seleet 15 samples of PLN as control group,and to select 62 samples of NHLas test group.Then accroding the different type of protein(P-gp,TopoⅡ,GST-π) andclinico-pathological data (sex character,age,IPI,LDH,β-2GM,B/T,Ann Arbor stage,clinical response)grouped.(2)All samples'fresh lymph nodes tissue were obtained from lymph nodes biopsy orneedle puncture and prepared suspended single cells by maehine separation.(3)We developed multiparameter reverse transcriptase-polymerase chainreaction(RT-PCR) method to semi-quantitate expression of P-gpmRNA,TopoⅡmRNA,GST-πmRNA of 62 non-Hodgkin'lymphoma, whileβ-actin as controlled within.(4)χ~2analysis and Spearman analysis were used to study the correlation of themultidrug resistance proteins with general clinical parameters and treatment outcome.Results (l) The expression of P-gp,TopoⅡand GST-πin NHL was 38.7%(24/62),50.0%(31/62)and 54.8%(34/62)respectively,compared with the control,the difference hasstatistically significance(P<0.05).(2) We did not found any significant statistical correlation between expression of P-gpand TopoⅡ(χ~2=0.44;r=0.480,P>0.05),P-gp and GST-π(χ~2=0.17; r=0.377, P>0.05),TopoⅡand GST-π(χ~2=0.26; r=0.355, P>0.05).(3) We did not find any statistical significant correlation between expression of P-gp inclinico-pathological data( sex character,age,IPI,β-2GM,B/T),there was significantstatistical correlation between expression of P-gp in I/II stage andⅢ/Ⅳstage(χ~2=9.789,P<0.05)and abnormal of LDH (χ~2=6.427, P<0.05).We also did not find any statisticalsignificant correlation between expression of TopoⅡand GST-πin clinico-pathologicaldata( sex character,age,IPI,LDH,β-2GM,Ann Arbor stage),there was significant statisticalcorrelation between expression of TopoⅡand GST-πin different types(χ~2=4.769; 3.847,P<0.05).(4) There was significant statistical correlation between expression of P-gp,TopoⅡand GST-πin chemotherapy drug resistance group(SD+PD)and chemotherapy sensitivegroup(CR+PR)(χ~2=5.79; 4.13; 5.17, P<0.05).Conclusion i)P-gp,TopoⅡand GST-πmay be a crucial factor involved in primaryresistance of NHLs,thus it may be useful prediction of che-motherapeutic treatment and riskof relapse;ii)it may help us to choose chemistry drug,enhance the sensitive ofchemotherapy.
