Objective: Butylphthalide is approved by the Food and Drug Administration of China for the use of a new drug for ischemic stroke,the previous study found that a variety of dementia animal model of learning and memory disorders have improved.In this study,we further investigated the effect of butylphthalide on learning and memory impairment and the possible mechanism associated with the protection of energy metabolism in Alzheimer's disease model rats induced by A?25-35,which aims to explore the anti-AD effects and possible related mechanism of butylphthalide.Methods: Male SD rats were randomly divided into normal group,model group and butylphthalide group?10,30,100 mg / kg?,each group of not less than 8.The model group and butylphthalide group were treated with one-time intraventricular injection of A?25-35.The next day of Model preparation,Butylphthalide group were given different concentrations of drug gavage,once a day at the same time until the end of the trial.Five days after A?25-35 Intracerebroventricular injection,the effect of butylphthalide on the learning and memory ability of rats was observed by water maze.The changes of mitochondrial function in brain tissue were observed by ATP,the purity of mitochondria was determined by measuring the content of lactate dehydrogenase and the activity of cytochrome c oxidase in the mitochondria.The mitochondrial activity of the mitochondria was detected by using the kit.SPSS17.0 was used for statistical analysis.The significant level ? was 0.05.The variance analysis and LSD test were used to determine whether there was significant difference between the two groups.Results: Water maze test showed that the learning and memory ability and spatial cognitive function of the vehicle group was lower than that of the control group while the NBP groups were significantly improved?P < 0.05?,indicating that the one-time intraventricular injection of A?25-35 preparation of AD rat model is more successful to continue the follow-up experiment.The learning and memory impairment of the butylphthalide group was significantly improved,the results showed that butylphthalide could significantly improve the learning and memory ability of dementia rats.Compared with the control group,the level of ATP in the vehicle group was significantly decreased,cytochrome c oxidase,pyruvate dehydrogenase complex and ketoglutarate dehydrogenase activity were also significantly decreased.Compared with the model group,butylphthalide can significantly against A?25-35 induced dementia rats learning and memory ability and ATP levels,Mitochondrial respiratory chain complex IV,pyruvate dehydrogenase complex,and alpha-ketoglutarate dehydrogenase activity.Conclusion: Butylphthalide on A?25-35 induced dementia rats learning and memory ability has improved significantly,and can improve mitochondrial function,the improvement of mitochondrial function by butylphthalide may be related to the mitochondrial respiratory chain function and the damage to the tricarboxylic acid cycle,reducing the release of mitochondrial complex IV,thereby reducing the death of A? abnormal neurons.In summary,butylphthalide nerve cell protection and learning and memory improvement has a significant effect,can provide a new idea for the treatment of AD.
