Objective:(1) By analyzing the expression of Ang-1 and VEGF in placenta from normal pregnant women and patients with hypertensive disorder complicating pregnancy, to explore whether both plays a role in the pathogenesis and significance for monitoring of hypertensive disorder complicating pregnancy. And therefore provides the theoretic foundation for early diagnosis and prognosis of hypertensive disorder complicating pregnancy.(2) Birthweight of all newborns were registered in order to know the effect of hypertensive disorder complicating pregnancy on fetal development.Methods:A random collection of 120 maternal cases who were delivered in obstetrics of the first hospital of ShanXi medical university in March 2009 ~ March 2010, which include 90 patients with hypertensive disorder complicating pregnancy (30 gestational hypertension, 30 mild preeclampsia, 30 severe preeclampsia) and 30 normal pregnant women.â‘ Placenta tissues were taked after delivery of the placenta, fixed by formalin and then made into paraffin embedded section, The changes of placental tissue were observed by light microscopy with hematoxylin-easin staining and the expression of Ang-1 and VEGF in placental tissue were estimated by immunohistochemical technology;â‘¡The gestational ages and birthweight of all newborns were registered. Experimental data of statistic methods: comparison in groups were conducted with one-way ANOVA, binary comparison between groups were performed with Bonferroni method, the relationship were analyzed with Spearman rank correlation analysis, P <0.05 indicate significant difference.Results:(1) The hematoxylin-easin staining results showed that: In the normal pregnancy group, the placenta villi arranged in neat rows, the vascular endothelial cells were normal, spiral arteries in placental bed with a very enlarged lumen, its vessel wall was thinner and the syncytiotrophoblast nodules can be saw occasionally; in the HDCP group, the placenta villi were thin and small, the vascular endothelial cells were loss, the vessels of placental villus and spiral arteries in placental bed with a very small lumen,its vessel wall was thick and fibrinoid necrosis, the placental villous syncytiotrophoblasts nodules increased obviously.(2) The expressions of Ang-1 of placenta in normal pregnant, gestational hypertension, mind preeclampsia and severe preeclampsia groups were gradually increased, there were significant differences among them ( P < 0.05).(3) The expressions of VEGF of placenta in normal pregnant, gestational hypertension, mind preeclampsia and severe preeclampsia groups were gradually decreased, there were significant differences among them ( P < 0.05).(4) The expressions of Ang-1/VEGF of placenta in normal pregnant, gestational hypertension, mind preeclampsia and severe preeclampsia groups were gradually increased, there were significant differences among them ( P < 0.05).(5) The expression of Ang-1 was reversely correlated to that of VEGF in HDCP placenta ( r = -0.866, P < 0.05).(6) The birth weight in normal pregnant, gestational hypertension, mind preeclampsia and severe preeclampsia groups were gradually decreased, there were significant differences among them ( P < 0.05).Conclusion:(1) The changes of expressional levels of Ang-1 and VEGF in placental tissue are related to the occurrence and development of hypertensive disorder complicating pregnancy, and the expression of Ang-1 and VEGF had a negative correlationship;(2) Hypertensive disorder complicating pregnancy affect fetal growth and development, the more serious conditions of the hypertensive disorder complicating pregnancy, the greater impact on the fetus.
